The major objective of the paper is through experiment research to explore protection mechanism of Arrowhead (Sagittaria sagittifolia) with its counter effect against hepatotoxicity induced by combination of isoniazid (INH) and rifampicin (RFP) on the treatment of tuberculosis. The paper did some tests on oxidative damage of liver and drug hepatic metabolism.At the same time, in order to provide sufficient theoretical basis for further research on Arrowhead, its relevant literature have been combed in all ages, and appropriate analysis of medical value, edible value, and health care value have been made.First of all,further literature reviews about87items pertaining to both medicine and food released in March,2002by Ministry of Health on their protection for liver injury have been searched. Among these items, Arrowhead has not been listed, but as ingredient of medicine and food since ancient times, it is of high value.Compared with nutrient value of Chinese yam which is praised as essential herb for tonifying deficiency, on the aspect of multiple nutrient contents,those of Arrowhead are not inferior. Furthermore, to clarify the relationships among Wuyu, Arrowhead, and Waternut Corm in ancient works and give a theoretical support for subsequent relevant studies.By criticism of phonology, botany, culturology, and philology, the article believes that Wuyu in Han and Wei dynasties refers to Arrowhead.The reason for the name of Wuyu changed to Waternut Corm is mainly due to understanding of"Wu"character whose original meaning indicating bird has been transferred to color of black. Because of changes in different names, some effects pertaining to Arrowhead have been shifted onto Waternut Corm.What's worse, due to similar ecological environment and root character, toxicity or/and side effect in Iphigenia indica has been meant to Arrowhead.As far as food safety considered, it is necessary to set their relationships straight. It is said that Arrowhead has a great effect on clearing heat and tonifying with its rich nutritive components and great health-keeping value.Secondly, the paper is going to detect protection mechanism of drug induced liver injury through duplicating model of rat's hepatic injury caused by combination of INH and RFP. After successfully establishing the model,blood biochemical indicators, pathomorphology of hepatic tissue, degree of oxidation of liver tissue and influence on metabolism after medicine entering liver were observed on the intervention of Arrowhead against combination of INH and RFP leading to rat's liver injury. Results show that after administration of Arrowhead juice, contents of alanine aminotransferase (ALT) and aspartate amino transferase (AST) in rat's serum greatly reduced compared with those of model group, which are of remarkable difference (P<.01). HE staining of rat's liver indicates Arrowhead's role in improving pathological lesion of liver.What's more, from Arrowhead's protection on degree of hepatic tissue oxidation, activity of nitric oxide (NO) in liver tissue with Arrowhead juice group is lower than that of model group. The activity of NO in medium and high concentration groups with Arrowhead juice, shows significant difference (P<0.01). Activities of Superoxide Dismutase (SOD) and glutathione S transferase (GST) recovered faster than those of model group. There is a significant difference between high concentration group and model group (P<.01). Activities of malondialdehyde (MDA) after lipid preoxidation in three experimental group decrease obviously compared with that of model group. Experimental group with three kinds of concentration shows extremely different (P<.01).All these statistics suggest Arrowhead indeed is effective in its protection against oxidative damage of liver.Finally, result shows, Arrowhead's protection against hepatotoxity induced by INH and RFP in drug metabolism, content of cytochrome P450(CYP450) in rat's hepatic tissue administrated by Arrowhead juice is a bit lower than that of model group, but there is no significant difference.However, content of cytochrome P4502E1belonging to one of more than20isozymes of P450is greatly lower than that of model group.CYP4502E1in the low concentration and high concentration groups displays significant difference (P<0.05),which gives some hint that it is related with pathomechanism of liver injury and also Arrowhead juice as an intervention has some effect in protecting liver.To sum up, Arrowhead can decrease activities of ALT and AST in rat's serum, alleviate rat's hepatic injury and improve pathological lesions of liver, which indicate Arrowhead's remarkable protection against oxidative damage of liver. After administration of Arrowhead juice, content of CYP4502E1in rat's liver tissue greatly decreased, which suggests Arrowhead juice's intervention in drug metabolism.
