| The donkey(Equus asinus)belongs to Perissodactyla,equine,and genus.Our donkey has a long history and is one of the major donkey countries in the world.As a domestic animal,the donkey has the features of rough feeding,sufficient tolerance,and strong disease resistance.Its meat,milk,skin have a higher nutritional value.As China’s economic and social development has entered a new stage,the donkey industry has displayed many important functions such as meeting product consumption,carrying agriculture and animal husbandry cycles,promoting structural adjustment,and now a variety of important functions such as poverty alleviation and poverty alleviationalleviating poverty and poverty alleviation.Donkey chestnut appear in the forehead of the donkey on the keratinized dermal tissue,folklore,which can be used as donkey night walking eyes;foreign scholars believe that it is preserved during the evolution of a donkey degenerative skin tissue.As the only raw material of animal skin gelatin,it has yet to see the transcriptional and proteomic level of animal skin and eye tissues night reported.In this study,four Dezhou donkey for the study,were collected each chestnut and the surrounding skin donkey front legs,the first is to provide data support for functional analysis of night vision;In addition,we also hope to identify the gene expression profile and protein expression profile of donkey skin.Through comparison with the gene expression profiles and protein expression profiles of other livestock skins,the possible molecular basis for the function of donkey skin was initially described.The preliminary research results are as follows: 1.Donkey chestnut transcriptomics studyUsing high-throughput sequencing to obtain the donkey hide and donkey chestnut transcriptome data and bioinformatics analysis,compared with the reference genome to identify differentially expressed genes;the results are as follows: a A total of 54158 genes were expressed,of which 15600 were shared in the chestnut and the skin.Compared with the skin,the total number of genes expressed significantly in the chestnut was 10925,of which 5949 were down-regulated and 4976 were up-regulated..1267 unique genes expressed in donkey skin,1105 expression genes unique to chestnut.The differentially expressed genes of GO and pathway were enriched and analyzed.The results showed that the differentially expressed genes were mainly enriched in the catabolic pathways of vitamin B6 and tyrosine;the high expression of DDC and BCL2 in may be related to the deeper skin color of the chestnut surface.2.Donkey chestnut proteomicsProteomics analysis was carried out through the iTRAQ quantitative project.The results were as follows: 17734 peptides and 3851 proteins were identified from the peptide map of donkey skin donkey eyes.The distribution of peptide length ranged from 5 to 23 aa.Compared with donkey skin,there were 1592 up-regulated proteins and 954 down-regulated proteins in the chestnut.The GO annotation and KEGG pathway functional enrichment analysis were performed on the differential proteins.The results showed that the differential proteins were mainly enriched in 77 GO items and 24 metabolic pathways.The retinol metabolic pathways appearing in the chestnut may be visually related.3.Donkey chestnut transcriptome and proteomic analysisIn order to further study the differences between the chestnut and the surrounding skin,the molecular basis for the difference between the donkey skin and the chestnut was elaborated.We analyzed the transcriptome and proteome data of donkey skin and and found that the total number of differentially expressed genes was 6601.A total of 742 proteins were associated with the transcriptome,indicating that the chestnut may be a special donkey skin tissue.Further,through BLAST comparisons,12 characteristic peptides were identified in the skin of the donkey,and the collagen GATGPAGVR sequence could be used as a characteristic component of the identification of donkey gelatin in the future. |
