Drug-loaded nanaofibers as a drug delivery is widely used in drug transportation and drug release.Drug-loaded nanofibers were achieved much attention because of slow release in the process of dose,spinning collagen nanofiber mat was prepared by electrospinning technique.The EDC/NHS and citric acid system were used to cross-link the nanofiber mat,and then the structure and property of uncross-linked and cross-linked nanofiber mat were researched.The smooth nanofibers with the uniform diameter were obtained under the optimal electrospinning condition which was 9%(w/v)concentration,the 20 kv voltage,the 15 cm distance,the 0.8 ml/h extrusion rate.The obvious cross-linking network can be observed for cross-linked the nanofiber with the EDC/NHS and citric acid system by the FSEM.It can be deduced that the cross-link process didn’t change the three helical structure of collagen by FTIR result.The hydrophilic property didn’t change by water contact angle result.The characterization of the degradation of uncross-linked and cross-linked nanofiber and the drug release was researched by combining the Coomassie brilliant blue method and the ultraviolet spectrophotometer.Uncross-linked nanofiber degraded in 10 days and the cross-linked nanofiber degraded in 21 days,so the property of the water resistance was obviously improved.The drug-loaded collagen nanofibers mat were prepared under the optimal electrospinning condition.The nanofiber morphology and the dispersity of 5-FU drug in nanofibers were researched by the FSEM and TEM.Any 5-FU drug particles can’t be observed on the surface of the fibers or in the fibers.That phenomenon illustrates that the 5-FU is perfect compatible with collagen and was distributed in the collagen fibers with molecular level.The drug release of the uncross-linked and cross-linked nanofibers was Studied.The result showed the cross-linked drug-loaded nanofiber reduce the drug burst release obviously and the utilization ratio of drug improved 10%-30%.This cross-linked nanofiber is a hopeful drug carrier which can reduce the pain and inconvenient it.
