Macleaya microcarpa (Maxim)Fedde is a species from Macleaya R. Br., Papaveraceae sp.,its stems and leaves are traditionally used for treating.The main active ingredients are alkaloids. M. microcarpa is especially concerned because of its variety of biological activities,such as anti-bacterial, anti-parasitic and improve liver function and so on. HPLC quantification of Protopine?Allocryptopine?Sanguinarine?Chelerythrine from roots?stems?leaves and fruits of M. Microcarpa. To explore dynamic accumulation of changes of four alkaloids from M. Microcarpa at different growth stages. Furthermore, the preparation process study of high purity allocryptopine from M. microcarpa fruit to develop a set of simple, convenient, environmentally friendly, low cost, and can be a large-scale production of high purity allocryptopine preparation process. This experiment has achieved the following progress:(1) Established a simple and reliable HPLC detection methods, to four kinds of alkaloids as a reference basis, by comparing the four kinds of alkaloids content changes in different harvest period, combined with its output to determine the best harvesting of medicinal herbs Phase; by comparing the four kinds of changes in the content of alkaloids in different parts of the growth to determine the best harvesting of medicinal parts for efficient and rational use of resources was provided.(2) Extracted with acidic water, adjust pH, macroporous resin enrichment, activated carbon purification, recrystallization and so on, can be prepared more than 98% of high-purity products of other hidden base, the yield was 1.0%, extraction rate of 90% or more. The process is easy, low cost, suitable for industrial mass production.
