Cryopreservation is the most promising way for long-term storage of biological samples e.g.,cells and cellular structures,which is of great significance for both scientific research and clinic application.Along with the wide application of stem cells and other materials in clinical and scientific researches,cryopreservation faces a growing number of small volume objects,and traditional methods have been unable to meet the demand.This paper presents a new method for the treatment of cryoprotectants for micro-volume biomaterials: a membrane separation is constructed on microfluidic chip,and micro fluid mass transfer occurring on both sides of the membrane to achieve adding or removing cryoprotectant agents.The process is smooth and controllable,and avoid additional mechanical damage.Based on this technical scheme,a series of investigations were performed and illustrated in this paper:(1)The principle design and analysis of microfluidic chip is studied: Based on the principle of microfluidic and membrane separation,this paper designs a membrane microfluidic chip system,and then the flow and transmembrane mass transfer models in the chip system are established.We can get guidance on the design and application of experimental systems and the feasibility of this program from the numerical analysis which is about this system;(2)The flow and mass transfer in membrane microfluidic system is studied: This paper designs and builds a microfluidic cryoprotective treatment experiment system based on microfluidic and membrane separation technology.On the basis of the experimental platform of this system,the cell-free experiment was carried out and the effect of membrane material and flow rate on the removal of cryoprotectants was investigated.(3)The removal of red blood cells cryoprotectants agents based on membrane microfluidic method is studied: Membrane microfluidic systems are used to treat cryoprotectants in red blood cells.The system's security and processing efficiency are tested for red blood cells.The system is used to optimize the treatment of red blood cells to remove glycerol.The above studies show that the method proposed in this paper can be used for small volume of biological materials to achieve a smooth and controlled cryoprotectants agent treatment,and it can improve the efficiency of erythrocyte cryoprotectants treatment.This method takes less than 5 minutes to treat 0.5ml of the erythrocyte cryoprotectants,and it can avoid cell damage caused by the osmotic pressure imbalance,reducing the difficulty of manual operation.This method is expected to play a major role in clinical medicine and scientific research.
