In terms of analysis of the traditional micro vision, Object image under micro vision is often obtained by the human eye or specific analysis software of observing image manual focusing. With the magnification of microscope becoming bigger and bigger, the depth of field become shorter and shorter. So manual focusing can not guarantee Z to reach the best focusing position when the height of micro object changes in most cases operation process like the assembly of micro parts, micro cell operation, integrated circuit assembly requires to be completed automatically, for positioning the blurred image can only get a clear image by automatic focusing. So it is necessary to study micro vision automatic focusing method and system based on the image process in order to guarantee real-time and accuracy of the operation process. In this papers the following research is carried out on the automatic focusing method and system of the micro vision in a single cell analyzer:The method and system of automatic focusing in the single cell analyzer were studied, combined with the characteristics of auto focusing under micro vision and FPGA control system based on its own requirements, the control platform based on FPGA controller is developedBased on the research of fast focusing algorithm of image process, fast focusing algorithm of image process under micro vision is researched in depth. Aimed at the deficiency of the algorithm, the function expression of the deviation of the target from the fuzzy difference is derived by using focal model of optical imaging system, the function expression of the deviation of the target can mediate on the Z axis motor to achieve fast focusing. The experimental results show only requiring two images of different degrees of deviation of the target is able to estimate the actual deviation of the target object with higher accuracy in order to achieve rapid and effective focus.Focus searching algorithm of adaptive variable step length is improved. Focus searching algorithm of an adaptive variable step length based on image process is developed. When far away from the best focus position, the included angle between the straight line and the horizontal straight line which is composed of two points on the evaluation function of any clarity is small, increase step length of the focus to achieve a coarse focus; When in the vicinity of the best focus position, decrease step length of the focus to achieve a fine focus. The algorithm is based on the premise of keeping focus accuracy, rapid focus can be achieved by using the evaluation function of the gray time domain clarity(4) In order to very conveniently observed automatically fast focusing results under the microscope vision and control the underlying hardware. The development of micro vision software on PC platform, underlying hardware device driver and design and compilation of fast focusing algorithm is studied. The development of micro vision software based on VS2010 platform is completed, the software can realize the image processing under micro vision and control of the underlying hardware.
