Skeletal muscle accounts for 40% of body weight,it has an irreplaceable role in many ways,such as exercising,breathing,maintaining the movement,regulating the temperature,regulating the metabolism.Generation of the skeletal muscle is a series of complex and delicate process,mainly in four aspects,the activation of muscle stem cells,proliferation and differentiation of muscle cells,cell fusion and multi-core muscle fibers.The large amount of exercise can pr omote skeletal muscle to generate reactive oxygen species,and which can pro mote skeletal muscle to generate,but there is little evidence for this directly.This article explores the effect of 8-oxo G produced by the movement in C2C12 on the proliferation and differentiation.Firstly,we had better divide the mice into two groups.One is the Static group,another is Sporting Group,wh ich must exercise in the downhill.We control its movement speed at 16 m/mi n,the slope at-16 o,and the time for exercise is 100 minutes at first.Then 5minutes for the rest,lastly,we let the sporting mice exercise for 100 minutes Again.By immunohistochemical method,we can get the relation between exerc ise and oxidative damage.Based on C2C12 differentiation model,C2C12 can b e divided into experimental group and control group by using 8-oxo G.The cell state and numbers of all periods we need can be observed by microscope,Th e expression levels of Myo D,M-Cadherin,and Myo G in the m RNA also can be detected by experiments.The following conclusions can be drawn from the study:1.By Immunohistochemistry test,it was found that the skeletal muscle of the experimental group produced a large number of 8-oxo G,indicating that the skel etal muscle injury caused by heavy load movement was more serious.2.By counting cells we found that mitosis phase in the stimulation group incr eased significantly than without stimulation group(P<0.01)when cell density was 80%,which shows that 8-oxo G produce by movement can significantly pro mote cell proliferation.3.We do a survey on RT-PCR in the stimulation group and without stimulatio n group cells,Results showed that three muscle molecular Myo D ?M-Cadherin and Myo G expression was significantly increased in m RNA(P<0.05)when ad ding 8-oxo G to stimulation group,which indicates that 8-oxo G produced by th e movement has significant effect on cell differentiation in skeletal muscle.
