Objective 1: To develop an UPLC method to determin the contents of gallic acid,tyrosol,salidroside,gallic acid ethyl ester and p-coumaric acid in Rhodiola algida.2: To investigate serum pharmaco chemical research of bioactive fraction of R.algida aganist chronic hypoxia-induced pulmonary arterial hypertension.Methods 1: The chromatographic separation of the analytes was achieved by using Zorbax Eclipse plus C18 column(2.1×100 mm,1.8 ?m).The mobile phase consisted of 0.05 aqueous phosphoric acid and methanol.Analytes were separated using a linear gradient with a flow rate of 0.4 m L/min.The detection wavelength was set at 275 and 308 nm and the column temperature was set at 30?.2: The absorbed components in rat serum after oral administration of bioactive fraction of R.algida(250 mg/kg,0.5,1,2 and 4h)were detected using an ultra high performance liquid chromatography-quadrupole-exactive orbitrap mass spectrometry(UPLC-Q-Exactive Orbitrap)method.Results 1: The calibration curves of the gallic acid,tyrosol,salidroside,gallic acid ethyl ester and p-coumaric acid were linear over 1.0~100.0,1.0~100.0,1.0~100.0,0.1~60.0 and 0.1~40.0 ?g/m L concentration ranges,respectively.The RSD of the instrument precision was within 0.75%.The current assay was stable and reproducible in quantification of all analytes with RSD within 3.90% and 3.66%,respectively.The average recovery of all analytes was in a range from 98.89% to 107.22%,with an acceptable RSD of no more than 3.82%.2: 10 absorbed components(Salidroside,Crenulatin,Creoside I,Tyrosol,Creoside II,Rhodioloside E,2-phenylethyl-1-?-D-glucoside,Sachalinoside B,Rosin and Rhodiooctanoside)were identified in rat serum after oral administration of bioactive fraction of R.algida using UPLC-Q-Exactive Orbitrap method.Conclusions 1: The UHPLC method can be used for the determination of gallic acid,tyrosol,salidroside,gallic acid ethyl ester and p-coumaric acid in R.algida.2: 10 absorbed components were detected in rat serum after oral administration of bioactive fraction of R.algida using UPLC-Q-Exactive Orbitrap method.
