| Qiannianjian was contained in the "Compendium of Materia Medica supplements",it has the effect of removing rheumatism and strong muscles.The traditional use of wind cold and damp arthralgia while mainly for the treatment of rheumatoid arthritis in the modern clinical.The quality control method of the TLC and the determination of gas phase content has been reported by the Chinese Pharmacopoeia.And the quality control standards need to be further supplemented and improved.It has anti-inflammatory,analgesic and other pharmacological effects.However,the experimental study of rheumatoid arthritis(RA)has not been reported,and the effect substance and mechanism are not yet clear.The research of the pharmacodynamic substance and mechanism for treatment RA on qiannianjian.And do further improve of the quality control standard of Rhizoma homalomenae.In order to provide more data,the basis and the reference for the clinical treatment of RA,also laid the foundation for the further development of homalomena.Method :⑴ To establish a method for determination of Homalomena HPLC fingerprint and linalool content based on the high performance liquid chromatography.Evaluating the quality of 10 batches of the Rhizoma homalomenae using the established methods.⑵Research the anti-inflammatory and analgesic effects of different extracts from Rhizoma homalomenae medicinal herbs based on the mouse auricle swelling test,rat foot swelling experiment,mouse torsion body experiment and rat Von Frey ciliary pain test.Combination the effects on AA rats.Screening of the basis for drug effect.⑶We observed the immune organs and ankle joints in rats,determined the serum cytokines levels of IL-1β,IL-2,IL-6 and IL-10 in rats,and studied the expression of NF-κB/p65 inflammatory protein in the ankle joints of AA rats.In order to reveal the mechanism of RA treatment of Rhizoma homalomenae with the adjuvant arthritis animal model.Result:⑴ A method for determining the content of linalool in Chinese medicinal materials was established in this experiment.The method of specific properties,repetitive RSD,diurnal difference RSD,daytime difference RSD,sample stability RSD,and linalool stability RSD were 2.04%,1.22%,1.18%,4.47%,1.55% respectively,and the average recovery rate was 101%,which conforms to the standard of Pharmacopoeia.A method for the analysis of HPLC fingerprint of Chinese medicinal herbs with good specificity,good separation degree,precision RSD,stability RSD and repeatability RSD 2.56%,3% and 2.26% respectively,which conforms to the technical requirements of fingerprint chromatogram,confirmed 11 common peaks,identified 1 peaks,and peak 7 was the peak of protocatechuic acid.10 batches of the overall fingerprint Homalomena is similar,but there are certain differences.The similarity of two batches of medicinal materials was poor and the rest were from 0.887 to 0.988.The similarity of S1,S2,S3 and S7 to the control atlas was up to 0.95.⑵ The volatile oil components and qiannianjian water extraction and alcohol precipitation supernatant fraction had anti-inflammatory and analgesic effect.Also,the volatile oil and linalool can reduce the weight of the adjuvant arthritis model rats,also reduce foot swelling and pain.We believe that Homalomena volatile oil and linalool may be one of the material basis for the efficacy of the treatment of RA.⑶ The volatile oil and linalool can down regulate rat ankle NF-κB/p65 protein,down regulate the level of IL-1β,IL-6 and IL-10 and raise the level of IL-2.This may be one of the mechanisms of volatile oil and linalool in the treatment of RA.Conclusion: This paper established the HPLC fingerprint of Homalomena medicine and the method for determination of linalool by HPLC.Supplement and improve the standard of quality control of Rhizoma homalomenae from the two levels of content determination and fingerprint.The volatile oil and linalool could be material basis for the efficacy of treatment of RA,its downregulation of NF-kappa B/p65 protein and regulation of serum cytokines such as IL-1β,IL-2,IL-6 and IL-10,it may be one of its mechanisms of action for the treatment of RA. |
