Objective: Explore clinical and laboratory factors that influence serum AMH testing results,to provide standardized operation for the clinical detection of serum AMH and to improve the accuracy of clinical diagnosis and treatment of AMH.Methods: A total of 105 women aged 20-30 years with normal ovarian function who were diagnosed in the outpatient clinic of the General Hospital of Ningxia Medical University from November 2016 to March 2017 due to a male factor and excluded:(1)polycystic ovary syndrome(PCOS)and rheumatoid Arthritis patients;(2)have basic diseases,blood lipids,bilirubin,hemoglobin and blood concentration abnormalities;(3)3 months prior to enrollment in steroids or ovarian surgery.To detect serum AMH before and after oral contraceptive pretreatment,GnRH-a downregulation,and serum AMH at fasting,drinking,or feeding,non-hemolysis and hemolysis,different storage temperature and time,and in different positions on the microplate by indirect test and t-test.The difference of serum AMH test results was statistically significant,that is,different Whether the treatment will affect the objective of serum AMH test results.Results 1.The serum AMH increased 14 days after GnRH-a down-regulation(P <0.05).2.No significant difference was found in AMH after 1-month pretreatment with oral contraceptives(P> 0.05).3.Drinking water or eating had no significant effect on the detection of serum AMH.4.Hemolytic group serum AMH than non-hemolytic group(P <0.05).5.Serum samples stored at 4°C for 1 day had higher AMH values(P <0.05);at 4°C,serum AMH showed statistically significant increases on the 3rd day(P<0.05)Serum AMH stored at-20°C for 1 month increased significantly(P <0.05);serum AMH stored at-40°C increased significantly at 4 months(P <0.05);and stored at-80°C.The sample did not change within 6 months(P> 0.05).6.The serum AMH placed on the microplate from the center to the periphery showed an increasing trend,and the AMH value of the most peripheral serum sample increased significantly.Conclusion: 1.The serum AMH value after using GnRH-a down regulation was significantly higher,not as an objective measure of ovarian function;2.Using oral contraceptives for a month does not affect serum AMH detection;3.Eating does not affect the serum AMH test;4.To ensure good stability of serum AMH,the temperature and duration of serum sample storage are as follows: only 12 hours at room temperature;2 days at 4°C;2 weeks at-20°C and-40°C,respectively 3 months;-80°C cryogenic preservation for at least 6 months.5.Sample hemolysis has a significant effect on test results.6.Placing serum samples on the edge of a microtiter plate may affect the objectivity of the AMH assay.
