Studies On The Chemical Constituents And Bioactivities Of Lindera Setchuenensis Gamble And Jacaranda Cuspidifolia Mart.

This project aims to study the chemical constituents of Lindera setchuenensis Gamble and Jacaranda cuspidifolia Mart.,test the Chemical composition biological activity from Lindera setchuenensis Gamble and Jacaranda cuspidifolia Mart.,which will provide new clues for the development and comprehensive utilization of the resources for these two plants.In order to obtain the total extract from Lindera setchuenensis Gamble by using the method of diacolation extraction,petroleum ether?ethyl acetate?n-butanol were added with extraction.Research on the chemical composition of Lindera setchuenensis Gamble.by various column chromatography methods.,such as silica gel column chromatography,Sephadex LH-20,RP-C18,19 compounds were obtained from Lindera setchuenensis Gamble.Using ¹H NMR,¹³C NMR,HR-ESI-MS and other modern analytical methods and TLC thin layer identification,and reference literature,the chemical structure of 14 compounds were determined.They are:4-Hydroxybulbocapnine?1???-?-Dicentrine?2???±?-O-acetylassythicine?3??Demethylmacrosporine I?4??Dehydronantenine?5??Stigmasterol?6???-Daucosterol?7???-Sitosterol?8???+?-Acortatarinowin F?9??Blumenol A?10??3,3-diethylpentane?11??2-Tetradecen-13-ynoic acid,2-?2-oxopropyl?-,methyl ester?12??Kaempferol 3,7-O-?-L-dirhamnoside?13??3,4-Dihydroxybenzalacetone?14?.Compounds 1,3,9-14 were isolated from Lindera setchuenensis Gamble for the first time.With using steam distillation to extract Jacaranda cuspidifolia Mart of volatile oil,85peaks were analysed and tested by GC-MS.33 compounds were identified by contrasting with MS spectrometry database,which accounted for 71.21%of the total content of volatile oil from Jacaranda cuspidifolia Mart.The main ingredients are:Palmitic acid?31.36%?,?Z?–9,17-Octadecadienal?12.06%?,Ethyl palmitate?3.81%?,Perhydrofarnesyl acetone?2.07%?,?-Maaliene?1.88%?,Cedro?1.42%?,9,12-Octadecadienoic acid ethyl ester?1.42%?Compounds 1,2,3,9 were selected to test the antitumor activity of B16 melanoma cells,C6 glioma cells,and HepG2 hepatoma cells using MTT assay.The results showed that compound 2 had a general inhibitory effect on the growth of HepG2 and C6,and the IC₅₀was 50.67 and 46.96?M,respectively.The in vitro antimicrobial activities of the essential oil were evaluated by the disc diffusion method,and the inhibition zones against Escherichia coli,Staphylococcus aureus and Candida albicans were 7.10,8.20 and 7.25 mm,respectively.There was no obvious bacteriostatic effect on Pseudomonas aeruginosa.
