Objective:This study explored the expression of S100P in endometrioid adenocarcinoma and its relationship with the clinical features of patients,followed by the role of S100P on endometrial cancer cell line AN3CA proliferation ability and migration ability.Methods:(1)In the first part of the study,the expression level of S100P in 45 cases of endometrioid adenocarcinoma tissues and 8 cases of normal endometrial tissues were detected by immunohistochemistry and qQT-PCR,then we analyzed the expression of S100P protein in different age,surgical pathological staging and histologic classification was analyzed;(2)transient transfection were carried out to construct the cell line AN3CA with overexpression of S100P gene,Western blotting and qQT-PCR confirmed the successful construction of AN3CA cell line overexpressing S100P,the cells transient transfected with plasmid were treated with experimental group,and AN3CA cells without pcDNA3.1-S100P plasmid were used as control group;(3)the effect of S100P on proliferation ability and migration ability of AN3CA cells was further verified by MTT assays and wound healing assays.Results:(1)Immunohistochemistry showed that S100P protein was highly expressed in endometrioid adenocarcinoma tissues(?~2=10.46,P=0.001),and q QT-PCR results also showed that S100P mRNA was highly expressed in cancer tissues(t=6.373,P<0.05),and the expression of S100P protein in different histologic classification groups is different(?~2=4.51,P=0.034);(2)the AN3CA-S100P cells were successfully constructed and the differences were significant(P<0.05);(3)the results of MTT assays and wound healing assays showed that S100P could enhance the proliferation ability and migration ability of AN3CA Cells(P<0.05).Conclusion:S100P is highly expressed in endometrioid adenocarcinoma and can promote the proliferation ability and migration ability of endometrioid adenocarcinoma cells AN3CA.
