Study one: Expression of thioredoxin and its reductase-1 in oral squamous cell carcinoma and its clinicopathological significance.OBJECTIVE: To study the expression and clinical significance of thioredoxin(Trx)and thioredoxin reductase-1(Trx R1)in tongue squamous cell carcinoma.METHODS:Twenty-eight cases of tongue squamous cell carcinoma and 10 cases of normal epithelial tissues were collected.The expression levels of Trx and Trx R1 in two tissues were detected by SP immunohistochemical three-step method.The relationship between their expression and clinicopathological features was analyzed.RESULTS: The expression levels of Trx and Trx R1 in tongue squamous cell carcinoma tissues were significantly higher than those in adjacent normal epithelial tissues.Trx IOD values in cancer tissues and adjacent normal tissues were respectively(2.84±0.34)×108 and(3.91±2.997)×107(P<0.001),Trx R1 IOD was(1.88±0.29)×108 and(0.69±0.32)×107(P<0.05);Trx and Trx R1 protein expression levels were between tumor size and differentiation The differences in expression were statistically significant(P<0.05);the expression levels of Trx and Trx R1 were moderately correlated(correlation coefficient r=0.504,P<0.05),and the high and low expression groups of Trx and Trx R1 were expressed.The difference in survival was statistically significant(P<0.05).CONCLUSIONS: Thioredoxin and its reductase-1 are the targets of drug therapy for tongue squamous cell carcinoma and important clinical indicators for early diagnosis and tumor screening.Study two: Effects of ethaselen,a novel organoselenium compound,on proliferation,cycle,apoptosis and migration in human tongue squamous cell carcinoma cells.Objective: To investigate the effects of novel organic selenium compound Ethaselen(BBSKE)on the proliferation activity,cell cycle,apoptosis and cell migration of human tongue cancer CAL27 cell line in vitro.Methods: MTT was used to detect the inhibitory effect of different concentrations of BBSKE on the proliferation of CAL27 cells.The apoptosis of CAL27 cells was detected by Hoechst 33258 immunofluorescence staining.The apoptosis rate and cell cycle distribution of CAL27 cells was detected by Flow Cytometry,the effect of different concentrat Effects of ethaselen,a novel organose Effects of ethaselen,a novel organoseleniumcompound,on proliferation,cycle,apoptosis and migration in human tongue squamous cell carcinoma cells.lenium compound,on proliferation,cycle,apoptosis and migration in human tongue squamous cell carcinoma cells.ions of BBSKE on the migration of CAL27 cells was detected by Transwell chamber.Results: BBSKE could significantly inhibit the growth and proliferation of CAL27 cells(P<0.01),and had drug concentration-dependent and time-dependent effects.By Hoechst 33258 immunofluorescence staining,BBSKE could obviously induce the apoptosis and morphological changes of CAL27 cells.Annexin V-FITC / PI staining showed that compared with the control group,BBSKE could significantly increase the apoptosis rate of CAL27 cells(P<0.01)and cause cell cycle arrest at G2 / M phase(P < 0.01).The results of Transwell chamber showed that the number of migrated cells in CAL27 cells decreased with the increase of BBSKE concentration,while BBSKE significantly inhibited the migration of CAL27cells(P<0.01).Conclusions:BBSKE can significantly inhibit the proliferation of CAL27 cells.BBSKE can induce cell cycle arrest in G2 / M phase and inhibit cell migration,suggesting that BBSKE can be used as an alternative chemotherapy for chemotherapy in TSCC patients.This study provides the experimental basis for its clinical application.
