Objectives:To investigate the effect of nanosecond pulsed electric field(nsPEFs)on systemic immunity and T lymphocytes cell.Methods:C57BL/6 mice with B16 malignant melanoma were exposed to 200 pulses of 100 ns duration and 30 kV/cm.The mice were sacrificed three days later.T lymphocytes were extracted from the spleen.Cell viability was evaluated by the CCK-8 assay.CD3~+CD4~+T cells,CD3~+CD8~+T cells,regulatory T cells(Treg)and myeloid-derived suppressor cells(MDSCs)were analysed by flow cytometry.The TNF-?,IL-2,IL-0,TGF-?,IFN-?levels in the supernatants were assessed by ELISA.Results:After the nsPEFs treatment,the number of T lymphocytes increased.CD3~+CD4~+T cells changed from 48%to 51.2%.CD3~+CD8~+T lymphocytes increased from 39.6%to 40.4%,Treg cells decreased from 4.3%to 2.4%,and MDSCs decreased from 39.0% to 19.7%.In addition,the levels of TNF-?and IL-2 increased(P<0.05).However,the level of IL-10 decreased(P<0.05),and the levels of TGF-?and IFN-?remained stable(P>0.05).Conclusions:Tumour growth can be effectively inhibited in vivo by nsPEFs,which activates the immune responses,inflammatory cells accumulated and immune cytokines changed. |
