Objective:To study different expression of P2X7 receptor in normal cervical tissue,cervical procancerous tissue and cervical cancer tissue,and find its relationship with staging of cervical cancer,high risk of HPV and expression of Ki-67 and P16.Methods:The study selected paraffin specimen of cervical procancerous tissue and cervical cancer tissue from patients who underwent cervical biopsy,LEEP surgical treatment and surgical treatment because of cervical lesions during January 2013 to June 2015 in our hospital.The paraffin specimen of normal cervical tissues were collected from patients with total hysterectomy for hysteromyoma.The stages of cervical cancer patients were obtained according to Uterine Cancers Clinical Practice Guidelines by FIGO.All patients were divided into 3 groups,the cervical cancer group?n=73,mean age 46.53±9.42?;the premalignant lesion group?n=90,mean age 46.83±9.92?;the normal group?n=112,mean age47.24±9.80?.In the cancer group,there were 73 cases with squamous cell carcinoma,and all tissues were not radiotherapy or chemotherapy.The staging of cervical cancer was Ia1 5 cases?7%?,Ia2 1 case?1%?,Ib1 21 cases?29%?,Ib2 9 cases?12%?,IIa1 23 cases?32%?,IIa2 11cases?15%?,IIb 3 cases?4%?.Among cervical procancerous group,there were 47 cases?52%?of LSIL and 43 cases?48%?of HSIL.Immunohistochemistry was used for determination of P2X7,Ki-67 and P16.Using real-time PCR to determine the highest risk type of HPV for pre-cervical cancer tissues and cervical cancer tissues.The results were analyzed by SPSS 18.0 statistical software.Results:1.Immunohistochemistry and the scores both indicated that expression of P2X7 was lowest in cervical cancer group and significantly lower compared with other two groupings,P<0.05.Meanwhile,expression of P2X7 in cervical procancerous group was significantly lower compared with the normal group,P<0.05.2.Tissue of Ib stage had significantly lower expression of P2X7 compared with tissue with Ia stage,P<0.05,meanwhile tissue of IIb stage had significantly lower expression of P2X7 compared with tissue with IIa stage,P<0.05.However no significant difference was observed between the tissue of Ia1 and Ia2,Ib1 and Ib2 or IIa1 and IIa2.3.Expression of P2X7 in tissues with HPV CT value<30 was significantly lower than that with HPV CT value?30,P<0.05.4.In cervical cancer group,ratio of high risk HPV patients increased with the increase of cancer staging,as well as expression of Ki-67 and P16,however expression of P2X7decreased with the increase of cancer staging and the expression of Ki-67 and P16.Conclusion:1.Expression of P2X7 in tissues with cervical cancer was significantly lower than that with cervical procancerous and normal cervical.2.Expression of P2X7 decreased with the increase of cervical cancer staging.3.Expression of P2X7 was positive correlated with HPV CT values in tissues with cervical cancer and cervical procancerous.4.Expression of P2X7 was negative correlated with expression of Ki-67 and P16 in patients with cervical cancer.In summary,P2X7 may be a new target in the diagnosis and treatment of cervical cancer.
