Objective: To investigate the protective effect of physcion on ? cells apoptosis of type I diabetes mice and cytokine-induced RINm5 F apoptosis.Methods: 40 mice were randomly assigned into a control group,a diabetes model group,physcion treatment group(low dosage)and physcion treatment(high dosage).Mice in diabetes group and physcion treatment groups received intraperitoneal injection of STZ to induce diabetic mellitus.1 week after STZ injection,mice in the low dosage and high dosage received injection at 20 and 40 mg/kg/d for 4 weeks,while mice in control and diabetes groups received injection of physiological saline for 4 weeks.Serum levels of NO,inducible nitric oxide synthase(i NOS)and insulin levels were measured by enzyme linked immunosorbent assay method.TUNEL assay was used to detect apoptosis.Moreover,an in vitro model using RINm5 F cells was established to evaluate the protective effect of physcion on cytokine-induced cell apoptosis.Results: Compared with the diabetes model group,the serum levels of NO and i NOS were significantly reduced in physcion treatment group(high dosage)whereas the insulin levels were significantly higher in physcion treatment group(high dosage).Moreover,apoptosis of ? cells were significantly reduced in physcion treatment group(high dosage)compared with diabetes group.In addition,our results demonstrated that physcion prevents cytokine-induced apoptotic cell death and leads to significant NF-?B suppressive effects in pancreatic ?-cells.Conclusion: Physcion can reduce the i NOS activity and NO production,in addition to effectively decreasing ? cells apoptosis.Our results suggested that physcion is a potential therapeutic agent against T1 DM.
