Sensitive skin(SS)is a high response state of skin produced under the physiological or pathological conditions,which is characterized by the enhanced perception reaction to the surrounding environment or local factors.It is preferred to call the SS as Self perception of sensitive skin(SPSS),and one of the main causes of SPSS is the skin barrier damage.The barrier sebum is the main component to maintain stability stability of the skin barrier,and its contents and/or structures changes can affect the skin barrier.Lipidomics,a research subject for lipid metabolism,is used to the study on screening and mechanism ofpotential lipid biomarkers.The facial lipids of SPSS and normal(NS)women aged 18-25 in Beijing were studied by ipidomics methods and results are as follows:1.The potential lipid biomarkers with significant differences in tow groups were screened combaining the UPLC-Q-TOF-MS technology and PLS-DA analysis method.The samples were divided into four groups: NS 1,NS 2,SS 1 and SS 2.The NS 1 and SS have the most significant difference,and 11 kinds of potential lipid biomarkers were screened.Among them,8 kinds ofpotential lipid biomarkers were higher in group NS 1,including 3 kinds of very long chain Cer,4 kinds of glycerophospholipids and 1 kinds of very long chain FFA,and other 3 kinds were higher in SS2 group,including 1kinds of short chain FFA and 2 kinds of DG.Combined with the physiological functions of various lipids,it is speculated that the reduction of lipids to maintain structural stability and the increase of the lipids to aggravate skin inflammation skin inflammation are the causes of the skin barrier dysfunction in sensitive skin.2.The barrier injury cell model were built by using the OECD guidelines in SDS stimulated HaCaT cells to screen the potential lipid biomarkers,GlcCer(d18:1/16:0)and Cer(d18:0/16:0).The results show that the twopotential lipid biomarkers have significantly recovery and no protective effect in the modle.3.The effect of the two kinds ofpotential lipid biomarkers on the expression of 7 kinds of lipid related gene in the repair processing were studied.The results showed that the expression of 6 kinds of lipid related gene were significantly decreased as the cell barrier injury,only one had no significant changes;the expression of 5 kinds of lipid related gene were significantly up-regulated treated by GlcCer/Cer,one gene had no significant changes;In summary,the barrier damage can inhibit the expression of lipid related gene even increasing the barrier damage;GlcCer/Cer can recover the barrier injury by up-regulating the expression of lipid related gene.The potential lipid biomarkers between NS and SPSS groups were screened by lipidomics methods,and the recovery mechanism of thepotential lipid biomarkers in the barrier injury cell model were studied.The study provide a theoretical basis for the development of cosmetics for SPSS.
