Macular degeneration is the most prevalent disease worldwide.In China,macular degeneration is a major disease that affects the vision of the elderly.However,due to the uncertainty of the pathogenesis and the lack of treatment and drug development,the research direction mainly focuses on the heritability of the pathogenesis of the disease.According to the data,there are 115 genes in 59 loci related to macular degeneration.FGD6 was identified as a PCV associated gene and its mechanism was related to the over-expression of FGD6.However,the effect of knock-down this gene in the animal model is unclear.AMD is a kind of diease which has correlations with other diseases based on the genetics data,for AMD genes associated with about 900 kinds of human disease.Now,the treatment of senile macular degeneration is relatively limited.Most drugsonly can stable vision but fail to improve vision Therefore,it is urgent to perform drug discovery for AMD.According to the above problem,this paper used siRNA to knock-down Fgd6 gene expression in mice retina to investigate its function for retinal vasculogenesis and performed a drug screen for AMD based on the public databases.The main contents are:1.Using siRNA to interfere with the expression of Fgd6 gene in mouse retina,the effect of Fgd6 gene knockdown on retinal blood vessels was observed by fluorescein fundus angiography.2.Used GWAS,KEGG,DGIbd as well as the Clue databases to identify senile macular degeneration related genes’ drugs.
