Current surgical managements of anterior cruciate ligament(ACL)rupture still remains an enormous challenge in ACL regeneration due to the poor healing potential of ACL.It is widely believed that inadequate cell numbers in the articular cavity contribute most to the negative consequence.This study developed a new knitted silk-collagen sponge scaffold by incorporation of allogeneic LSPCs sheet to provide sufficient suitable seed cells and natural extracellular matrix.Excellent proliferation and mult-differentiation capacity ofLSPCs were proved in the initial stage of study.This study include two parts,ectopic implantation and orthotopic transplantation.In ectopic implantation,ACL treated with the silk-collagen sponge scaffold with LSPCs sheet showed less immune cells,more fibroblast-like cells and extracellular matrix.With a rabbit ACL injury model,better collagen fibril formation could be observed in orthotopic transplantation.After 6 months,the LSPCs sheet group had more mature ligamentous structure.At the same time,no obvious cartilage and meniscus degeneration were observed at 6 months post-operation.In conclusion,these results demonstrated that the silk-collagen sponge scaffold with LSPCs sheet can promote ACL regeneration and slow the progression of osteoarthritis,suggesting its bright prospect in ACL reconstruction clinic use as an ideal graft.
