Objective:In this experiment,we studied the effects of iRoot BP Plus bioactivity and SP on bone formation in blank control subgroup,iRoot BP Plus subgroup,iRoot BP Plus+SP subgroup,and the quantity and number of inflammatory cells in bone defect.For the improvement of iRoot BP Plus,the corresponding experimental basis is provided to provide more support for bone loss caused by chronic periapical periodontitis and chronic periodontitis.Method:32 SD female rats were randomly divided into 4 groups,8 rats in each group.At the top of the cranial top of each rat,three 1.5mm deep and 2mm diameter of round hole bone defects were made,each defect was seperated by 2mm.From front to back,three bone defects were placed blood clots,iRoot BP Plus,iRoot BP Plus+SP respetively to establish a blank control subgroup,iRoot BP Plus subgroup and iRoot BP Plus+ SP subgroup bone defect model.At 2,4,8,12 weeks after operation,each group sacrificed 8 rats by using overdose of anesthetic drugs and then cranial sections was obtained.HE staining and Masson staining were made to observe the number of inflammatory cells.Histologic observation and quantitative analysis were used to evaluate the quality and quantity of new bone formation.Result:The inflammatory reaction was present at the four intervals for all the tested materials.4 week interval had the most severe inflammatory response,but at 12 week interval,the reaction was minimal in all cases.Also,a dissolution reaction was observed for all the materials.At 2 week interval and 4 week interval,the inflammatory reaction of iRoot BP Plus subgroup and iRoot BP Plus+SP subgroup were lighter than that of the blank control subgroup(P<0.05).At 4 week interval and8 week interval,compared with the blank control subgroup,the new bone formation in the iRoot BP Plus subgroup and i Root BP Plus+SP subgroup were more(P<0.05).At 4 week interval compared with i Root BP Plus subgroup,new bone formation in iRoot BP Plus+ SP subgroup was more(P<0.05).Conclusion:(1)From the results of new bone formation,it is concluded that neuropeptide P can promote new bone formation in a certain period of time.(2)The following conclusions can be drawn from the inflammatory bone defect classification: iRoot BP Plus,iRoot BP Plus+SP has a good inflammatory regulation and osteogenic potential.
