Purpose:(1)To investigate the expression of MSI2 protein in colorectal cancer and its relationship with clinicopathological features Explore the expression of MSI2 protein in colorectal cancer.(2)To investigate the effect of MSI2 on the proliferation and invasion of colon cancer cell lines.Method:(1)Select 153 paraffin-embedded specimens and 35 para-cancer paraffin specimens from patients with colorectal cancer(CRC)who were operated on from November 2009 to November 2011 in Department of Gastrointestinal Surgery,the First Affiliated Hospital of Nanchang University.Immunohistochemistry(IHC)was used to detect the expression of MSI2 in 153 specimens of cancer tissues and 35specimens of adjacent tissues.Western blot was used to demonstrate the retesting of MSI2 expression in 4 fresh CRCs and paired paracancer tissues and colon cancer cell lines.(2)Statistical software was used to analyze the expression of MSC and MSI2 in CRC specimens and paracancerous tissues,and the extent of tumor invasion,metastasis of lymph nodes,distant metastases,Dukes stage,tumor marker CEA,age of patients,patient gender,CRC size,location of tumor,tumor classification,and tumor differentiation.(3)Construct plasmid vectors and siRNAs to determine the proliferation,clone,migration,and invasion ability of MSI2 overexpressing and MSI2 silenced colon cancer cell lines by in vitro cell assay.Result:(1)IHC results showed that the expression of MSI2 in CRC specimens was significantly higher than that in paracancerous tissues and was mainly expressed in the cytoplasm.Western blot results also confirmed that the expression of MSI2 in CRC specimens and colon cancer cell lines was significantly higher.(2)There were54 cases(54/153,35.3%)with high expression in CRC tissues,and 4 cases(4/35,11.4%)with high expression in tissues adjacent to tumors.The X~2test analysis results showed that X~2 was 16.09.P value<0.001,the difference was statistically significant.High expression of MSI2 protein in CRC and have positive correlation with tumor infiltration depth,lymph node metastasis,distant metastasis and Dukes stage.However,the high expression of MSI2 protein in CRC have no significant difference in patient age,patient gender,tumor size,location of tumor,tumor classification,tumor differentiation and tumor marker CEA.(3)In vitro cell experiments showed that determining the overexpression of MSI2 can promote the proliferation,clonality,and migration and invasion ability of colon cancer cell lines.Conversely,silencing the expression of MSI2 will play an inhibitory role.Conclusion:The expression of MSI2 was elevated in CRC tissues and in each colon cancer cell line,and the site was mainly in the cytoplasm.(2)The expression of MSI2 was related to the depth of tumor invasion,lymph node metastasis,presence or absence of distant metastasis and Dukes stage.However,it has nothing to do with the patient's age,gender,tumor size,tumor location,tumor type,tumor differentiation and tumor marker CEA.(3)The expression of MSI2 can promote the proliferation,clonality,migration and invasion of colon cancer cells.
