ObjectiveWe develop a model in vitro to study the differentiation of BMSCs by co-culture with lung tissue of COPD model mouse?Fig 1?and explore the induced differentiation of BMSCs by co-culture with the suspension of lung tissue of COPD model mouse..Method:The Co-culture experiments were carried out in the transwell model.Experiment group A:momoplast suspension of lung tissue of COPD model mouse and P3BMSCs;Experiment group B:momoplast suspension of lung tissue of normal model mouse and P3BMSCs;Control group:P3BMSCs were cultured separately.After ten days,we were going to measure the expression of SP-C and AQP-5 in BMSCs via Western blotting and RT-PCR.Result:1.There were wheezing in the COPD mouse model,the lung tissue expanded and were less elastic.There were impression when pressing into the surface with a finger.HE staining showed that the brochial mucosal epithelium exfoliated,with a large number of inflammatory cells infiltrated.the alveolar spaces got larger,the alveolar walls were thickened,even broken.and the walls of the vessels were thickened.the semi-quantitative analysis showed the MAN in the COPD muose model was significantly less than the healthy control group,while the MLI was significantly larger than that of the healthy control group?P<0.01?.2.In the experiment group A,the morphology of BMSCs was gradually changed from spindled-shape to paving-stone shape and analogous to epithelial cell.In the experiment group B and Control group,the morphology of BMSCs still presented with long spindle-shaped.3.In the experiment group A,the expression of AQP-5,SP-C protein and mRNA of BMSCs were both significantly higher than that in the experiment group B and control group?P<0.01?.4.With the co-culture time prolonged,The expression of AQP-5,SP-C protein and mRNA of BMSCs in the experiment group A showed an increased tendency,the difference was statistically significant?P<0.01?.Conclusion:1.The mouse model of COPD was sucessfully established by passive smoking and intratracheal instillation of lipopolysaccaride.2.In this experiment,We established experiment model about momoplast suspension of lung tissue of COPD model mouse and BMSCs3.The differentiation of BMSCs into alveolar epithelial cells can be induced by lung tissue of COPD model mouse.
