Objective:To investigate the method of using artificial human venous blood to simulate arterial thrombosis in vitro and the experimental study on the evaluation of physical characteristics.Method:Venous blood was taken from healthy blood a donor that was per person 18ml,According to the different methods of thrombus preparation,they are divided into 4 groups:venous blood?group A?,enous blood+CaCl2?group B?,venous blood+CaCl2+thrombin?group C?,venous blood+CaCl2+thrombin+ADP?group D?.12thrombi per group,each thrombus weighs about 120.2mg±2.9mg.Intracoronary thrombus in acute myocardial infarction patients within 2 hours as control group?Group E?.Thrombus in each group compared with E group structural components and evaluation of its performance.Percutaneous Coronary Artery Implantation?PCI?was used to implant the self-made mixed thrombus into t the distal end of the left anterior descending coronary artery?LAD?of Beagle to prepare an acute myocardial infarction model.Result:Pathological results,compared with E group:only the main pathological structure of D group mixed thrombus is consistent with E group,that is,HE results showed that only a large number of irregular platelet-stained platelet trabeculae were observed in group D;Masson results showed a large number of irregular,continuous,blue-stained fibrin formations;SEM results showed,compared with groups A and B,group C produced a relatively increased number of fibrin and a loose fiber network formed by their interweaving,showing a large number of red blood cells and several activated platelets;in groups A and B,Only fine fibrin was connected between blood cells,but no fibrous network was formed.In the in vitro thrombolysis assay,Group D rate of lysis was highes,with significant differences between groups?P<0.001?;The Beagle AMI model was successfully prepared after placing the self-made mixed thrombus into the target vessel.Conclusion:The venous blood+ADP+thrombin+CaCl2 group is most likely to form mixed thrombus withplatelets and fibrous reticular structures,which has a better stability,higher cracking rate,thus,it can provide a stable and simple thrombus model for clinical thrombolysis experiments.
