Objective:The aim of present study was to investigate the protective effect of vitamin U on renal toxicity induced by valproate sodium(VPA)and provide laboratory data for clinical application of VPA.Methods:In this study,48 female rats were used and they were 6~6.5 month(mean weight150~190g).These animals were randomly divided into 4 groups: control group(group A),vitamin U group(group B),valproate group(group C),vitamin U+ sodium valproate group(group D).These rats was fixed daily at same time.Group A was given the same amount of normal saline(gavage),group B was given VitU 50mg/kg/d(gavage),group C was given VPA300mg/kg/d(gavage)and group D was given VitU50mg/kg/d firstly,then VPA300mg/kg/d after 1 hours.Weigh the weight per week and observe the weight changes of these animals.Treated for 2W or 4W,then not eating for one night and were anaesthesiaed.Total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL),low density lipoprotein(LDL),Serum creatinine,urea and uric acid were detected by automatic biochemical analyzer.The renal tissue was observed histopathologically.Result:1.Influence on blood lipidThere was significant differences in TC and LDL between the group A and group C(P>0.05),and the level of TC and LDL in group C were significantly higher.2.Influence on serum biochemical indexes of renal functionThere was no significant difference in creatinine,uric acid and urea between four groups at 2W(p>0.05).At 4W,compared with the other three groups,the creatinine,urea and uric acid level of VPA group was significantly higher and the difference was statistically significant(58.67±5.55mmol/l,10.34±1.13?mol/l,80.55±12.13 mmol/l).But there was significant difference among the group C and the group D,and the levels of creatinine,urea and uric acid in group D were significantlyreduce.3.Influence on pathological morphology of renal tissueAt 2W,there were no obvious abnormality in renal structures among the four groups.4W,Inflammatory lesions were only seen in VPA group,and mild inflammatory cell infiltration were seen in other three groups.Conclusion:VPA can lead to a higher level of blood lipid.The renal toxicity induced by VPA may have a certain relationship with the time of drug exposure,and vitamin U has a protective effect on the renal toxicity induced by sodium valproate.
