Human adipose derived stem cells(hADSCs)are promising candidates in regenerative medicine.But as serial passages during cell culture,a gradual decline of fitness occurred,such as loss of abilities of proliferation and differentiation,and etc.Since PQQ is a famous antioxidant,cofactor of dehydrogenase and amine oxidase,we examined possible anti-senescence effects of PQQ on secreted factors of hADSCs.hADSCs from donors were cultivated,long-term passaged until proliferation arrest occurred.Various concentration of PQQ(1.5,3.0,6.0,9.0,15,30.0 nm/ml)were added into the culture medium.We collected culture supernatant treated with or without PQQ,found that PQQ down-regulated mRNA expression of IL-1?,TGF?-1PDGF,up-regulated mRNA expression of IGF-1,bFGF and PDGF.ELISA results revealed significant increase of IGF-1 and bFGF mRNA expression after treated with PQQ for 72 h.Cell proliferation was measured by CCK8,senescence associated?-galactosidase expression were evaluated after 72 hours,results suggested that PQQ had obvious effect on hADSCs proliferation,while ROS contents were decreased.Therefore PQQ may have protective effect on hADSCs from UVA irradiation through secret paracrine factors.
