ObjectiveObesity is a worldwide metabolic syndrome,the number of obesity increased year by year.To explore the mechanism of obesity has important theoretical significance meaning.Recent researchesfind that Erythropoietin(EPO)can be used to treat obesity,but the mechanism is not that clear.Obesity causes hypoxia in white adipose tissue,and the expression of hypoxia inducible factor 1?(HIF1?)increases in tissues.The composition of adipocyte extracellular matrix contains rich collagen,collagen type I(COL1)andcollagen type VI(COL6)are the most abundant.Obesity leads to the development of adipose tissue fibrosis,and transforming growth factor ?1(TGF?1)is reported to be involved in the formation of adipose tissue fibrosis.In this study,obese mice induced by high fat as an obesity model to explore the EPO mechanism of improvement of obesity,with a view to provide clues for the treatment of obesity.Methods1.Comparation of adipocytes in adipose tissue between Con mice and EPO mice by HE staining and Masson stainingParaffin sections stained with HE and Masson,photographed using an ordinary microscope.2.The total mRNA inthe subcutaneous fat of the control group and the EPO group was extracted separatelyandthe level of HIF1?,TGF?1,COL1 and COL6 mRNA was detected by Real-time PCR.3.The total protein in the subcutaneous fat of the control group and the EPO group was extracted separately,and the level of HIF1?,TGF?1,COL1 and COL6 protein was detected by Western blot.Results1.Compared with CON group,he body weight of the EPO group was reduced(31.05 + 0.82 g vs.23.48 + 0.82 g)(P<0.001),the content of subcutaneous fat mass(0.840 + 0.013 g vs.0.720 + 0.018 g)(P<0.01)andthe content of epididymal fat masswere reduced(0.870±0.018 gvs.0.590±0.019g)(P<0.001).Food intake of the two group is no significantly different.2.Compare with CON group,the level of blood triglyceride has no significantly different in EPO group,the level of blood glucose decreased(87.90+2.44 mg/dl vs.61.80+ 1.47 mg/dl)(P<0.001).3.Compared with CON group,the size of subcutaneous adipocytes decreased and the fibrosis of subcutaneous adipose tissue improved to a great extent.4.Compared with CON group,the level of HIF1? mRNA in EPO group decreased(1.09+0.04 vs.0.87+0.02)(P<0.01),the level ofTGF?1 mRNA in EPO group decreased(0.710 ± 0.021 vs.0.610 ± 0.018)(P < 0.01),the level of COL1 mRNA in EPO groupdecreased(1.007±0.035 vs.0.920±0.025)(P<0.05),and the level of COL6 mRNA in EPO group decreased(1.09±0.04 vs.0.87±0.02)(P<0.01).5.Compared with CON group,the level of HIF1? protein in EPO group decreased(0.590±0.014 vs.0.450±0.015)(P<0.01),the level ofTGF?1 protein in EPO group decreased(0.31±0.01 vs.0.24±0.01)(P<0.01),the level of COL1 protein in EPO group decreased(0.510±0.012 vs.0.380±0.011)(P<0.05),and the level of COL6 protein in EPO group decreased(0.680±0.015 vs.0.540±0.017)(P<0.001).ConclusionEPO can significantly reduced the body weight of high fat-induced obese mice and reduce their blood glucose,improve the collagen hyperplasia obese mouse model.
