BackgroundPM2.5 refers to the aerodynamic diameter less than or equal to 2.5 microns of atmospheric particulates,due to its small particle size,able to get the body with the air alveoli,so it is also known as can go directly to the alveoli.In recent years,frequent fog weather,a serious threat to people's life quality and health,has become the attention of the government,the public and researchers.Epidemiology and toxicology,etc.The study found that exposure to PM2.5 environment is closely related to the incidence of thyroid cancer,this study adopts the different concentrations of PM2.5 in the canister in rats,observe the PM2.5 in rat thyroid PI3K/AKT signaling pathway.ObjectiveApplication of different concentrations of PM2.5 solution to infected rats,observe the of rat thyroid tumor related PI3K/AKT signaling pathway and the effect of explore possible mechanism of thyroid lesions,preparation for the prevention and treatment of thyroid disease.MethodCollection between December 2016 and March 2016 discovery of PM2.5 samples,made different concentration 0.9% saline suspension.Set aside.Level 60 healthy clean male Wistar rats were randomly divided into 5 groups: normal saline control group,0.75mg/kg dose group and 2.5mg/kg dose group,the 5mg/kg dose group,10mg/kg dose groups,each group of 12.Using noninvasive inhaled tracheal instillation of groups of rats infected,according to 0.5ml/kg dose,canister to 3 times a week,canister to eight weeks in a row.The control group with normal saline instead of particles suspension.At the end of the last infected the next line of femoral artery bleeding to death.Quickly remove the thyroid,part of the specimen in 4% paraformaldehyde fixed,the rest of the specimen in a-80 refrigerator.? Immunohistochemical determination of p-PI3K?p-AKT?cyclinD1 and Bcl-2?the amount of p53 protein expression;Real-time fluorescent quantitative PCR detection of PI3K?AKT?cyclinD1 and Bcl-2,p53 gene expression;Western Blot detection p-PI3K?p-AKT?cyclinD1 and Bcl-2,the amount of p53 protein expression.Result1?Compared with saline control group,0.75 mg/kg dose group of PI3 K,and AKT,cyclinD1,Bcl-2 gene variations of no statistical significance(P>0.05),2.5 mg/kg dose group,5 mg/kg dose group,10 mg/kg dose group of PI3 K,and AKT gene expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with the dose of 0.75mg/k group,2.5mg/kg dose group,5mg/kg dose group,10mg/kg dose group of PI3 K,and AKT,cyclinD1 and Bcl-2 gene expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with 2.5mg/kg dose group,5mg/kg dose groups,10mg/kg dose group of PI3 K,and AKT,cyclinD1 and Bcl-2 gene expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with 5mg/kg dose group,10mg/kg dose group of PI3 K,and AKT,cyclinD1,Bcl-2 gene expression differences of no statistical significance(P>0.05).2?Compared with saline control group,0.75mg/kg dose group of p53 gene variations of no statistical significance(P>0.05),2.5mg/kg dose group,5mg/kg dose group,10mg/kg dose group,the expression of p53 gene significantly lower amount,the difference was statistically significant(P<0.05);Compared with the dose of 0.75mg/kg group,2.5mg/kg dose group,5mg/kg dose group,10 mg/kg dose group,the expression of p53 gene significantly lower amount,the difference was statistically significant(P<0.05);Compared with 2.5mg/kg dose group,5mg/kg dose groups,10 mg/kg dose group,the expression of p53 gene significantly lower amount,the difference was statistically significant(P<0.05);Compared with 5mg/kg dose group,10mg/kg dose group of p53 gene expression differences of no statistical significance(P>0.05).3?Compared with saline control group,0.75mg/kg dose group p-PI3K? p-AKT?cyclinD1,the change of the Bcl-2 protein has no statistically significant difference(P>0.05),2.5mg/kg dose group,5mg/kg dose group,10mg/kg dose group of p-PI3 K,and p-AKT,cyclinD1,the Bcl-2 protein expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with the dose of 0.75mg/kg group,2.5mg/kg dose group,5mg/kg dose group,10mg/kg dose group of p-PI3 K,p-AKT,cyclinD1 and Bcl-2 protein expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with 2.5 mg/kg dose group,5mg/kg dose group,10mg/kg dose group p-PI3 K,p-AKT,cyclinD1 and Bcl-2 protein expression quantity increased significantly,the difference was statistically significant(P<0.05);Compared with 5mg/kg dose group,10mg/kg dose group of p-PI3 K,p-AKT,cyclinD1 and Bcl-2 protein expression differences of no statistical significance(P>0.05).4?Compared with saline control group,0.75mg/kg dose group of p53 protein variations of no statistical significance(P>0.05),2.5mg/kg dose group,5mg/kg dose group,10 mg/kg significantly lower dose group,the expression of p53 protein content,the difference was statistically significant(P<0.05);Compared with the dose of 0.75 mg/k group,2.5mg/kg dose group,5mg/kg dose group,10mg/kg significantly lower dose group,the expression of p53 protein content,the difference was statistically significant(P<0.05);Compared with 2.5mg/kg dose group,5mg/kg dose groups,10mg/kg significantly lower dose group,the expression of p53 protein content,the difference was statistically significant(P<0.05);Compared with 5mg/kg dose group,10mg/kg dose group of p53 protein expression differences of no statistical significance(P>0.05).ConclusionPI3K/AKT signaling pathways involved in regulating the PM2.5 of thyroid of rats caused by injuries.
