Objective:To observe the effect of chiropractic method on the inflammatory response of chondrocytes and the signal transduction pathway of p38MAPK in cervical spondylosis model rats.Methods:SD rats were used for a total of 40 males and females.Each group consisted of 10 animals,divided into sham operation group,model group,model massage group(abbreviated as Tuina group)and mobicor group,for a total of 4 groups.The model group referenced the cervical disc degeneration model;the sham group only cut the skin and then sutured it as a blank control.The preparation model was completed and kept for 90 days for staining(toluidine blue).The standard of success:changes in the morphological structure of rat intervertebral discs after modeling.After successful modeling,it was divided into 3 groups of 8 in each group.They were:model control group(referred to as model group),model chiropractic group(chiropractic group),and model mobital group(abbreviated as mobicon group).Chiropractic group:Self-made elastic band method for secure fixation.The experiment was as follows:the rat was fixed with one hand and the skin of both sides of the spine was lifted up by the thumb pulp of the other hand and the pulp of the index finger.From the base of the tail to the "Ojodet",the movement was performed once and repeated alternately.The total operation was 14 times.From the 8th time,the flexor lateral edge of both hands was used to hold the back of the rat's back.The index finger was pressed forward with the middle finger,and the four fingers were lifted at the same time.The back skin of the rat was lifted from the tail base to the "Dazhui".Using the method of pinching three times;one time a day,one course of two weeks,interval two days,continue the following course of treatment,a total of two courses.Mobi can be grouped:intragastric intervention,the amount of mobectin tablet is 0.7878 mg/kg a day,the same time in the same group;model group,sham group:model group and sham group,no treatment,more than the same pinch The ridge group is consistent.At the end of the treatment,toluidine blue was used to stain the tissue.Immunohistochemistry was used to detect the content of IL-6 and IL-8.Western Blot was used to detect the expression levels of MKK3/6 and p38MAPK.To investigate the effects of Chiropractic method on chondrocyte inflammatory factors and its effect on p38MAPK signaling pathway,the anti-inflammatory effect of chiropractic method was proposed,which may be mediated by p38MAPK signaling pathway.Results:(1)Toluidine blue staining:sham group:The number of cartilage endplates is large,dark blue-purple,thick and the extracellular matrix is abundant.Model group:The cartilage endplates were thinner,lighter in staining,and less in number and extracellular matrix.(2)Immunohistochemistry(IHC):Compared with the sham-operated group,the positive expression levels of IL-6 and IL-8 in the model group were significantly increased,and the difference was statistically significant(P<0.05);compared with the model group,In the Chiropractic group and the Mocombe group,the protein expression levels of IL-6 and IL-8 were all significantly reduced,and the differences were statistically significant(P<0.05).(3)WestemBlot test:Compared with the sham group,in the model group,the positive expression of MKK3/6 and p38MAPK protein was significantly increased(P<0.05);compared with the model group,the chiropractic group The expression levels of MKK3/6 and 38MAPK were significantly decreased in the mobibe group,and the difference was statistically significant(P<0.05).Conclusions:(1)Reduce the expression of IL-6 and IL-8 protein in intervertebral disc chondrocytes by chiropractic therapy.(2)Down-regulate the protein expression of MKK3/6 and p38MAPK in the cartilage of the intervertebral disc by chiropractic therapy.(3)Inhibiting inflammatory reaction of the cartilage endplate and degeneration of the intervertebral disc by chiropractic therapy.This mechanism may be down-regulated the positive expression of IL-6 and IL-8,probably due to the regulation of the MKK3/6-p38MAPK signal transduction pathway.
