ObjectiveTo investigate the inhibitory effects of celastrol on invasion and migration of ovarian cancer SKOV-3 cells and its mechanism,in order to provide theoretical basis for celastrol intreatment of ovarian cancer.Methods1.MTT assay were used to measure proliferation of cells and select the appropriate concentration of celastrol.2.Transwell chamber experiments and scratch repair test were used to test invasion and migration of SKOV-3 cells.3.Western blot was used to detect the expressions of EMT-related proteins E-Cadherin,Vimentin,Fib,Snail and TGF-?/Smad signaling pathway protein p-Smad2 and p-Smad3.Results1.MTT showed that celastrol inhibited the proliferation of SKOV-3 cells in a time and dose dependent manner.2.Transwell chamber experiments showed that the cells invasion rate was significantly increased after TGF-?1 induction,while the cells invasion rate was decreased in a dose-dependent manner after celastrol treatment(P<0.05).3.The scratch test showed that after the induction of TGF-?1 cell migration rate was significantly increased,and the cell migration rate of celastrol decreased after celastrol treatment,in a dose-dependent manner(P<0.05).4.Western blot results showed that compared with the blank group,the expression of E-candherin in TGF-?1 group was down-regulated and the expressions of Vimentin,Fib and Snail were up-regulated,inducing the occurence of EMT.Compared with TGF-?1 group,in TGF-?1+celastrol(0.5?M,0.75?M,1.0?M)group,the expression of E-Candherin was up-regulated,and the expression of Vimentin,Fib and Snail was down-regulated,reversing the occurence of EMT.Meanwhile,the expression of p-Smad2 and p-Smad3 in TGF-?/Smad signaling pathway was also down-regulated(P<0.05).ConclusionCelastrol can inhibit migration invasion and migration of ovarian cancer SKOV-3cells,which may reverse the occurrence of EMT in SKOV-3 by inhibiting TGF-?/Smad signaling pathway.
