ObjectiveApoE-/-mice induced by atherosclerotic diet were subjected to vibrational exercise intervention to observe the effect of vibrational exercise on atherosclerosis in mice and its possible mechanism.Methods1.32 healthy 8-week-old ApoE-/-mice were randomly divided into 2 groups:normal diet group?16?,atherosclerosis diet group?16?,given normal and atherosclerosis,respectively Feed for 8 weeks.Eight weeks later,the mice in the normal diet group were randomly divided into two groups:normal control group?NC group,8 rats?,normal vibration group?NV group,8 rats?,AS group were randomly divided into two groups:AS control group AC group,8 rats),AS vibration group?AV group,8 rats?.Vibrating intervention was performed on the two groups of mice in NV group and AV group.The vibrational modes were:frequency 15 Hz,peak amplitude 2.0 mm,time 30 min,time 1,day 6,week 12.2.After 2 and 12 weeks of shaking test,weight of mice in each group was weighed,serum HDL and LDL were measured by enzyme method,serum ox-LDL and VLDL were detected by ELISA,and observed by oil red O staining,HE staining,and Massion staining.Atherosclerotic plaque surface area,foam cells and collagen fibers.Western Blot was used to detect the semi-quantitative expression of TLR4,MyD88,NF-?B,IL-1 and IL-6 in the aortic TLR4/NF-?B signaling pathway.Results1.Body weight and blood lipid of ApoE-/-mice in each group:There was no significant difference in body weight between the groups after 12 weeks of vibrational exercise.Serum HDL levels in AC group were significantly higher than those in NC group?P<0.05?.There was no significant increase in LDL,VLDL,and ox-LDL.The difference was not statistically significant.HDL was significantly increased in NV group.The difference was statistically significant?P<0.05?,LDL and VLDL were significantly decreased,the difference was statistically significant?P<0.05?,and there was no significant change in ox-LDL.The difference was not statistically significant.Compared with AC group,serum LDL,VLDL and ox-LDL in AV group also decreased significantly,the difference was statistically significant?P<0.01?,HDL had no significant change,the difference was not statistically significant.2.ApoE-/-mice aorta oil red O staining results:Compared with the NC group,aortic plaques were significantly increased in the AC group,and the difference was statistically significant?P<0.05?.NV aortic plaque reduction,but no significant difference.Compared with AC group,AV aortic plaque reduction,this difference was statistically significant?P<0.05?.3.Compared with NC group,the number of aortic foam cells in AC group increased significantly,the number of necrotic crystals in plaque increased,and the aortic intima increased obviously.NV group aortic intima more smooth,only protruding.Compared with the AC group,the aortic intima-media foam cells in the AV group were reduced,the intimal thickening was reduced,and the necrotic core was reduced.4.ApoE-/-mouse aorta Massion staining results:Compared with NC group,collagen fibers of aortic plaque in AC group decreased.The number of collagen fibers in the NV group increased.Compared with the AC group,collagen fibers in the aortic plaques of the AV group increased.5.The expression of TLR4/NF-?B related protein in mouse aorta by Western Blot showed that compared with NC group,expression of TLR4,MyD88,IL-6 in aortic tissue of AC group was significantly increased,the difference was statistically significant?P<0.01,P<0.001?.There was no significant difference in NF-?B and IL-1 between the two groups.The level of NF-?B in NV group was significantly decreased?P<0.001?,while there was no significant difference in TLR4,MyD88,IL-1 and IL-6.Compared with AC group,the expression of TLR4,NF-?B,MyD88,IL-1 and IL-6 in the aortic tissue of AV group mice was significantly lower?P<0.01,P<0.001?.ConclusionsVibration exercise can reduce the serum LDL,VLDL,ox-LDL and aortic plaque area.Vibrational motility delays the progression of atherosclerosis by inhibiting the TLR4/NF-?B signaling pathway.
