ObjectiveTo investigate the protective effect of Astragalus polysaccharide(APS)on cardiomyocyte apoptosis induced by lipopolysaccharide(LPS)in mice and its mechanism.MethodsIn the cell experiment,H9c2 cardiomyocytes were preincubated with high,medium and low concentrations of Astragalus Polysaccharide(10 mg / L ~ 25mg/ L ~ 50 mg /L)for a total of 24 h after incubation with LPS(1 UM)for a total of 24 h to establish a cardiomyocyte apoptosis model.Then MTT assay was used to detect the activity of cardiomyocytes.The activity of cardiomyocytes was detected by MTT assay.The expression of I?B-?(cytoplasm),NF-?B(cytoplasm),NF-?B(nuclear),p-JNK and JNK were detected by Western blot.In animal experiment,two doses of astragalus polysaccharide(200 mg /kg,400 mg / kg,800 mg / kg)were given preventively by SPF Kunming mice,and then LPS(10 mg / kg)was injected intraperitoneally to establish myocardial apoptosis model of myocardial cells.Cardiac function index of cardiac ejection fraction(EFSs),left ventricular shortening fraction(FSs)and left ventricular shortening fraction(FSs)were measured by echocardiography 8 h later.;And then put the animals to death,take the heart,.TUNEL assay was used to measure the apoptosis rate of cardiomyocytes and ELISA was used to detect the protein content of IL-1 ? and TNF-? in serum.Western blot was used to detectthe expression of I ? B-?(cytoplasmic fibronec? B)protein,and Bcl-2 family,caspase-3 related protein expression.ResultsThe MTT assay showed that the activity of cardiomyocytes in LPS model group decreased significantly,while the activity of cardiomyocytes was significantly increased compared with model group after APS intervention.Compared with the control group,LPS model group can significantly inhibit left ventricular systolic function in mice,TUNEL test results showed that apoptotic cardiomyocytes were significantly increased;ELISA results showed that serum IL-1?,TNF-? increased;Western Bcl-2 decreased.The levels of NF-?B and I?B-? in the cytoplasm decreased while the concentrations of NF-?B in the nucleus increased.Astragalus polysaccharide pretreatment group compared with LPS model group,can significantly inhibit LPS-induced left ventricular contraction in mice decreased,the relative decrease in apoptotic cardiomyocytes;The expression of P-JNK,Bax and Caspase-3 in the serum of IL-1?,TNF-? and myocardial tissue decreased,while the expression of Bcl-2 increased but the expression of JNK did not change.-?protein expression increased,while the nuclear NF-?B decreased.ConclusionsAstragalus polysaccharide can protect LPS-induced cardiomyocyte apoptosis in vitro and in vivo,which may be related to the inhibition of NF-?B and JNK signaling pathway.
