ObjectiveTo observe the effects of resveratrol on anti-Müllerian hormone,luteinizing hormone,follicle-stimulatinghormone,estradiol,malondialdehydeand superoxide dismutase in experimental rats with premature ovarian insufficiency?POI?and the expression of PI3K,p-PI3K,Akt,p-Akt,mTOR,p-mTOR,Bax,Bcl-2 and Caspase-3 in ovarian tissue,and explore the mechanism of resveratrol.action.MethodsA total of 90 5-week-old SPF grade SD healthy female rats were randomly divided into low dose resveratrol group?18 rats in group a?and middle dose resveratrol group?18 rats in group b?high dose resveratrol group?18 rats in group c?,model control group?18 rats in group d?,blank control group?18 rats in group e?.The successful model rats?72 rats given tripterygium glycosides tablets 50mg/?kg·d?for 14 days according to the observation of indicators to determine the premature ovarian insufficiency?were randomly divided into low-dose treatment group,middle-dose treatment group,high-dose treatment group and model control group.The low-dose,middle-dose and high-dose resveratrol treatment groups were given intragastric doses of 20mg/kg,40mg/kg and 80mg/kg daily,while the model control group was orally administered with 0.5%sodium carboxymethyl cellulose solution.The levels of serum sex hormones:anti-Mullerian hormone?AMH?,luteinizing hormone?LH?,follicle stimulating hormone?FSH?,estradiol?E2?and serum malondialdehyde?MDA?and superoxide dismutase?SOD?levels were measured by ELISA.Follicles,atresia follicles and luteal number.The expression of p-Akt,p-mTOR,Bax,Bcl-2and Caspase-3 in ovarian tissue were compared by immunohistochemistry.The expression of PI3K,p-PI3K,Akt,p-Akt,mTOR,p-mTOR,Bax,Bcl-2 and Caspase-3 in ovarian tissue were detected by Western-blot.ResultsThe serum levels of AMH,E2 and SOD in rats of different concentrations of resveratrol in treatment group were higher than those in model control group?P<0.05?;while the levels of LH,FSH and MDA were lower?P<0.05?;the expression of p-PI3K,p-Akt,p-mTOR and Bcl-2 in ovarian tissue was high?P<0.05?;while the expression of Bax and Caspase-3 was low in ovarian tissue?P<0.05?.The levels of serum AMH,E2 and SOD in serum of rats with different concentrations of resveratrol were significantly lower than those of the blank control group?P<0.05?,while the levels of LH,FSH and MDA were higher?P<0.05?;ovarian tissue expression of p-PI3K,p-Akt,p-mTOR,Bcl-2 protein low?P<0.05?,Bax,Caspase-3 protein expression high?P<0.05?.Compared with the low-dose treatment group,the middle-dose treatment group,the high-dose treatment group and the blank control group,the number of granulosa cells layers,the number of mature follicles and the number of corpus luteum in the model control group were decreased?P<0.05?,and the number of atresia follicles was significantly increased?P<0.05?.Conclusions1.Serum levels of AMH,E2 and SOD in rats with premature ovarian insufficiency after intragastric administration of resveratrol were increased,the levels of LH,FSH and MDA were decreased;the number of granulosa cell layers in ovary,the number of mature follicles and Luteal number is increased,the number of atresia follicles significantly decreased.Ovarian function in rats with premature ovarian insufficiency has been improved.2.The expression of p-PI3K,p-Akt,p-mTOR and Bcl-2 in ovarian tissues of rats with premature ovarian insufficiency after intragastric administration of resveratrol was rose,and the expression of Bax and Caspase-3 was reduced.Resveratrol activates the PI3K/Akt/mTOR signaling pathway and inhibits granulosa cell apoptosis.3.Resveratrol on the treatment of premature ovarian insufficiency rats by activating PI3K/Akt/m TOR signaling pathway and anti-oxidative stress mechanism,play a role in inhibiting granulosa cell apoptosis.
