ObjectiveBreast cancer resistance potion(BCRP)and the ubiquitinated E3-linkedenzyme Bre1 A are closely related to the occurrence and development of malignant tumors.However,the mechanisms by which they regulate the occurrence and development of laryngeal carcinoma are not yet clear.In order to investigate the effects of BCRP and Bre1 A on the proliferation and migration of laryngeal carcinoma cells,we investigated the function and role they played in the development of laryngeal carcinoma.Provide valuable theoretical basis for the future clinical diagnosis and treatment.MethodsWe used PCR to amplify the human breast cancer resistance genes BCRP and Bre1 A from the mammary gland library and correctly inserted them into Myc and 2B-Flag vector.The recombinant plasmids and empty vector were transfected into 293 T cells respectively.The expression of Hep-2 was detected by Western blot,and the growth curve and cell scratch test were performed.ResultsWe successfully cloned human BCRP size of about 1835 bp,human Bre1 size of about 2927 bp,PCR products were successfully inserted into the Myc and 2B-Flag tag by sequencing to prove the successful construction of Myc-BCRP,2B-Flag-Bre1 A,and western blot detection Myc-BCRP,2B-Flag-Bre1 A fusion protein.The results showed that after 293 T cells hadtransfected with the recombinant plasmids Myc-BCRP and 2B-Flag-Bre1 A,obviously specific bands were observed at a relative molecular weight of about73 KD and a relative molecular weight of about 120 KD visible obvious specific bands,indicating that the recombinant proteins Myc-BCRP and 2B-Flag-Bre1 A can be correctly expressed in 293 T cells.Growth curve was used to detect the effect of BCRP and Bre1 A on the growth of laryngeal carcinoma cells(Hep-2).The results showed that BCRP could promote the growth of laryngeal carcinoma cells,while Bre1 A could inhibit the growth of laryngeal carcinoma cells.Cell scratch assay was used to detect the effects of BCRP and Bre1 A on laryngeal carcinoma cells.The results were consistent with the growth curve.ConclusionsBCRP promotes the proliferation and migration of laryngeal cancer cells,and Bre1 A inhibits the proliferation and migration of laryngeal cancer cells.This study has further explored the development of laryngeal cancer for BCRP and Bre1 A,providing a new theoretical basis for the follow-up study of the occurrence and development of laryngeal cancer.
