Hydroxytyrosol(HT),which is found in primary olive oil,is of high medical value.It is considered to be one of the representative substances of olive oil to play antioxidation.Previously,foreign scholars have studied the pharmacological effects of HT on anti inflammation,antithrombotic,blood lipid regulation,anti arteriosclerosis,retinal macular degeneration,protection of cartilage and anti osteoporosis.Some studies have also discussed the pharmacological mechanism of HT at the molecular level.In recent years,more and more attention has been paid to the anti-tumor effect of HT.It is believed that HT can play a role in the treatment of cancer by inhibiting the formation of blood vessels,and the results show that HT can play a role in different stages of angiogenesis and have multiple targets,and can change the cell distribution of the proliferation cycle of the epithelial cells.At the same time,it has been reported that HT has a preventive effect on retinal macular degeneration by preventing the oxidative damage of retinal epithelial cells by preventing acrolein,environmental toxicity,oxidation of fat particles and age.Retinal macular degeneration is mainly manifested in the decline in the phagocytosis and digestion of retinal epithelia,secondary to subretinal neovascularization,blood vessel leakage and bleeding,and a series of secondary pathological changes.Its prevalence increases with age.It is an important disease in the middle-aged and the elderly.So far,in modern medicine,no effective and effective drug therapy has been found to prevent the progression of the disease.In order to prevent and treat macular degeneration,it is necessary to protect retinal epithelial cells from oxidative damage and to maintain their ability to swallow and digest.On the other hand,it needs to inhibit the neovascularization of the retina.From these two points,compared with the biological activity of HT,if HT can effectively inhibit angiogenesis,it is likely to be a new therapeutic drug for retinopathy.At present,the research on HT in China mainly focuses on health products and food additives,and there are few studies on its medicinal aspects.However,from the point of view of various biological activities of HT,the possibility of HT is increasing continuously with the in-depth study of its pharmacological effects.Therefore,we want to explore the effect of HT on retinal neovascularization,and to explore the mechanism of its action to provide a possibility for the treatment of retinopathy.In this study,we study the OIR model,CAM model and EA.hy926 vascular endothelial cells in two levels in vivo and in vitro,and discuss the effect and mechanism of HT on retinal angiogenesis,and further develop the medicinal value of HT,and provide a possibility treatment scheme for retinal neovascularization diseases.First,in vitro,EA.hy926 cells were intervened with different concentrations of HT.CCK-8,cell scratch and Matrigel methods were used to observe the cell proliferation,migration and the formation of vascular endothelial cells,so as to explore the effects of HT on endothelial cells,and the Western Blot method was used to detect VEGF eggs.The expression level of white was used to preliminarily study the mechanism of HT affecting endothelial cell growth.Then the CAM model was constructed to observe whether HT inhibits angiogenesis induced by Bfgf.Finally,the Oxygen-induced retinopathy(OIR)model was established,and the pharmacodynamics of HT was evaluated through the methods of vitreous injection and the methods of retina paving and HE staining.The results of CCK-8,cell scratch and Matrigel in the external environment were summarized and summarized.It was found that HT could inhibit the proliferation of EA.hy926 in vascular endothelial cells,inhibit the normal migration of the cells,and inhibit the endemic endovascular structure of vascular endothelial cells.Western Blot method was used to determine the expression level of VEGF.The results showed that HT could inhibit the expression of VEGF protein and had a certain concentration dependence.Therefore,the inhibitory effect of HT on angiogenesis may be related to the down-regulation of VEGF expression.Using CAM vascular model to investigate the effect of HT on angiogenesis,it can be observed that HT can inhibit angiogenesis induced by Bfgf.In vivo,OIR was successfully constructed and vitreous injection was administered to HT.The results of HE staining showed that HT could reduce the number of nuclei breaking through the inner boundary membrane of the retina;the results of immunohistochemistry showed that HT could reduce the expression level of VEGF;and the results of Western Blot and RT-PCR showed that HT was probably down regulated by the expression of HIF-1 alpha and VEGF,thus inhibiting the angiogenesis of retinal vascularization.In general,this topic is to discuss the effect of hydroxytymol on retinal angiogenesis.The OIR model,CAM model and EA.hy926 vascular endothelial cells are selected as the research models,and a series of experiments have been carried out.It is found that HT can inhibit retinal angiogenesis at two research levels in vitro.In addition,its mechanism of action was preliminarily studied,and its inhibitory effect was related to down regulating the expression of HIF-1 and VEGF.It provides some experimental evidence for the future study of anti angiogenesis effect of HT,and provides a possible treatment for retinal neovascularization.
