Mesenchymal stem cells(MSCs)are considered as ideal seed cells and one of the reasons for their therapeutic effect in regenerative medicine is their powerful immunomodulatory ability.The rigidity of different tissues in vivo are quite different.It has been found that the rigidity of the matrix can regulate the differentiation direction of MSCs,but the regulation of the immunomodulatory ability of MSCs is deficient of report.The purpose of this project is to investigate the alteration of immunomodulatory ability of MSCs on matrix of different rigid.To achieve this purpose,the co-culture of MSCs with PHA-induced lymphocyte proliferation was used as an in vitro model to track their immunosuppressive capacity.The lymphocyte proliferation was detected by flow cytometry using CFSE labeling.The gene expression of immune-related molecules by MSCs were further analyzed.The results showed that the immunoinhibitory ability of MSCs was weakened in the GelMA group of low rigidity.The gene expression of immunomodulatory molecules(IDO,iNOS,TSG-6,TGF-?,PGE2,HGF,etc.)were higher in the GelMA group of low rigidity,while the gene expression of MHC-I and co-stimulatory molecules significantly decreased.Besides,the gene expression of adhension molecules and chemokines were also affected by GelMA rigidity,but there were no consistency.This study expands our knowledge about the immunobiology of MSCs on matrix of different rigidity.
