Objective: to study the beneficial effects of Fecal microbiota transplantation and Fecal microbiota transplantation combine with pectin on DSS-induced colitis mice in weight,DAI activity,Inflammatory cytokine,stool character,histological changes of colon,zonula occludes protein,peripheral monocyte subtype and comparing the proportion of M1/ M2 macrophages in mucosa and submucosa.and study the influence on intestinal microflora and its metabolites of short chain fatty acids.To explore the possible mechanism of Fecal microbiota transplantation for ulcerative colitis.Methods: C57BL/6 mice were given 3.0%DSS for 7 days to induce colitis,and then randomly divided into DSS-FMT group(colitis model group,6 rats),DSS+FMT group(FMT group,6 rats),DSS+FMTP group(FMT combine with pectin,6 rats),NC group(control group,6 rats)given distilled water feeding.Subsequently,Corresponding groups were treated with fecal filtrate enema and gavage.The body weight,diet,mental state,fecal property and disease activity index(DAI)were observed daily.Fifteenth days,mice were killed and samples of colon,serum and feces were collected.Immunohistochemical staining,immunofluorescence,HE staining,flow cytometry,enzyme-linked immunosorbent assay,16 SrRNA high throughput sequencing,gas chromatography and other technical means were used to study the related detection indexes.Results: Group DSS+FMT,group DSS+FMTP and group DSS-FMT had significant difference in weight(17.05 ± 0.70 g,17.45 ± 0.50 g,vs 16.03 ±0.23g)at day 10(P < 0.05);On day 12,Group DSS-FMT DAI index(3 ± 0.63)was significantly higher than that in group DSS+FMT(2.33 ±0.52)and group DSS+FMTP(1.67 ± 0.52)(P < 0.05);In group DSS-FMT,the colon length(5.78 ± 0.38cm)was significantly shorter than that of group DSS+FMT(6.69 ± 0.50cm)and DSS+FMTP group(7.03 ± 0.47cm)(P < 0.05).At the same time,compared with the DSS-FMT group,the DSS+FMT group decreased the inflammatory cytokine IL-6(53.28±4.40 pg/ml vs 75.35±6.78 pg/ml)?TNF-?(221.76±15.31 ng/L vs 273.10±21.20 ng/L),IL-1?(63.81±7.01ng/L vs 76.87±5.86ng/L);improved anti-inflammatory cytokine IL-4(37.19±7.86 pg/ml vs 16.08±12.33 pg/ml)?IL-10(50.24±10.66pg/ml vs 33.13±11.24pg/ml)(P<0.05);It also improves the histologic damage and changes the proportion of Ly6C+/Ly6C-in peripheral blood mononuclear cells.from the DSS-FMT group 62.17±6.24%/ 37.83±3.45% to 45.68±3.79%/43.23±5.86% in DSS+FMT group.The M1/M2 distribution and proportion of macrophages in intestinal mucosa were also affected.It increased the diversity of the intestinal flora,the abundance of beneficial bacteria,increased the level of many short chain fatty acids in the intestine,and the fecal bacteria transplantation combined with pectin treatment showed a more positive effect.Conclusion: 1.Fecal microbiota transplantation and Fecal microbiota transplantation combine with pectin treatment can improve the general condition of the colitis mice(mental state,eating and drinking water,hair,activity,stool characteristics).It can also improve the degree of weight loss in the colitis mice,the activity index of DAI disease,the shortening of the colon length,the histopathological injury and restores the expression of tight connexin in the intestinal mucosa.to promote the balance of inflammatory cytokines and anti-inflammatory cytokines in the colon tissue.2.Fecal microbiota transplantation can increase the abundance and diversity of intestinal flora in colitis mice,and to a certain extent,promote the transformation of abnormal intestinal flora to normal flora.At the same time,It can increase beneficial microbiota producing short chain fatty acids,inhibit harmful bacteria and improve the content of various short chain fatty acids in the intestinal tract of colitis mice,and play a positive role in adjusting the flora.3.The mechanism of the positive effect of Fecal microbiota transplantation for colitis may be related to the abnormal immunization of bacteria,which may involve the mononuclear macrophage system.
