Background Clinically,spinal cord injury is a serious central nervous system injury and it is the most serious complication along with spinal injury.The reasons are traffic accidents,altitude crashes and sports injuries.In addition,after long-term chronic fatigue of the spine and overwork,there will be the spinal degenerationthe,disc herniation,yellow ligament hyperplasia calcification and so on.It will give oppression to the corresponding stage of spinal cord tissue,blood supply is limited,caused intramedullary signal changes,Spinal cord cells will degeneration and necrosis,followed by abnormal skin and limbs and limb dysfunction.Different stages of spinal cord injury,the clinical symptoms are not the same.Overall it is characterized by high fatality and high disability,and the clinical intervention is limited,often leads to severe limb dysfunction in the injury phase.Polymeric inositol polycytiridin(Poly-IC)is a ligand for Toll-Like Receptor 3(TLR3),the study found that its preconditioning has a protective effect on ischemic encephalopathy and renal reperfusion injury,it can reduce the infarct size,reduce tissue edema,improve neurological function score and reduce the occurrence of immune inflammatory reaction.Thus,according to the mechanism of Poly-IC,we propose to use a model of SCIRI.We intent to investigae the effect of Poly-IC on apoptosis,edema and expression of related inflammatory factors after spinal cord ischemia-reperfusion injury in rats.To confirm whether it plays a protective role after spinal cord ischemia-reperfusion injury and provide a new basis for further study on the medicine used for SCIRI.Objective We propose to use a model of SCIRI on rats,Ischemia-reperfusion group were administered after modeling intravenous injection of Poly-IC.To further esplore the impact of TLR3 ligand Poly-IC on tissue edema,inflammatory factor and apoptosis in rat spinal cord ischemia-reperfusion injury,to investigate it is protective effect after spinal cord ischemia-reperfusion injury.Methods A total of 72 healthy adult male SD rats were randomly divided into sham-operation group,ischemia-reperfusion group and Poly-IC group,and reperfusion 60 min later.Sham-operation group only opened abdominal cavity,rats models of spinal cord ischemia-reperfusion injury were established by clamping the abdominal aorta in the later two groups.Rats in ischemia-reperfusion group and Poly-IC group were injected with 0.1ml of saline and 1.25?g/g Poly-IC through abdominal cavity after model establishment.BBB scores were test at 6h/24h/48 h in each group after modeling success,ELISA were used to test TNF? and IL-10;Immunohistochemistry were used to test NF-?B and IL-10 at 48 hours,we used TTC staining and HE to observe spinal cord tissue morphology change.Results Each group for 48 h,Compared with ischemia-reperfusion group(3.8±0.75),BBB scores were significantly improved(P<0.01)in Poly-IC group(9.4±0.49),with time progressed gradually,functional activities of limbs gradually recovered,HE staining showed pathological injury of spinal cord tissue significantly reduced,TTC staining showed that the area of ischemic necrosis of spinal cord tissue was significantly reduced,the expressions of NF-?B were reduced and IL-10 were increased in immunohistochemistry.Each group for 48 h,Enzyme-linked immunoassay showed IFN?(55.65±4.02),TNF?(201.82±2.18)?IL-1?(50.59±1.47)in ischemia-reperfusion group,compared with this,the content of IFN?(117.23±6.06)was increased(P<0.01)in Poly-IC group,the contented of TNF?(190.45±4.16)was reduced(P<0.01)and the contented of IL-1?(39.27±2.48)was reduced(P<0.01).It shows that the application of Poly-IC after spinal cord ischemia-reperfusion injury can relieve spinal cord injury and improve functional status of limbs.Conclusions Poly-IC can reduce the inflammatory factors after spinal cord ischemia-reperfusion injury in rats,reduce the inflammatory response,reduce tissue edema,it has a protective effect on the spinal cord.
