Objective:To observe the effect after electroacupuncture(EA)in Inflammatory Bowel Diseases(IBD)mice,the number of defecation times in unit time of model mice?small intestinal propulsive rate and colon length?and changes in the expression of different adenosine receptors in the colon;To evaluate the effect of electroacupuncture sensitized acupoints on intestinal motility in mice with inflammatory bowel disease,and explore the possible mechanisms,so as to provide laboratory evidence for clinical treatment of inflammatory bowel disease by electroacupuncture.Methods: 48 male SPF C57 mice were randomly divided into four groups,the normal group,the model group,the EA sensitized acupoint[“Dachangshu”(BL25)] group and the EA non-sensitized acupoint group[ “ Xinshu ”(BL15)],12 rats in each group.The inflammatory bowel disease model was established by TNBS(2,4,6 three nitrobenzene sulfonic acid ethanol)method.EA sensitized acupoint group,EA non-sensitizatized acupoint group were treated with electroacupuncture at “ Dachangshu ”(sensitized acupoint)and “Xinshu”(non sensitized acupoint).The time of electroacupuncture was 30 minutes,and it was treated continuously for 7 days.No treatment was used in the normal group and the model group.To observe the disease activity index(DAI)of IBD model mice after modeling?the number of defecation in 2h,to measure small intestinal propulsive rate after electroacupuncture treatment and changes in the length of the colon,Western blot(WB)was used to detect the expression of A1 and A3 receptors in the colon of the model mice.Result: 1.Compared with the normal group,the EA sensitizatized acupoint group and the EA non-sensitized acupoint group after modeling,the DAI and the number of defecation in 2h of model group increased significantly,the difference was statistically significant(P < 0.05).2.The DAI and the number of defecation in 2h of EA sensitized acupoint group and EA non-sensitized acupoint group after first,fourth,seventh days of EA treatment was gradually decreased as the number of days increased(P < 0.05).And also the effect of EA sensitized point group is more obvious than EA non-sensitized acupoint group(P < 0.05).There was no significant difference in the number of defecation in 2h in the normal group and the model group with the number of days increased.3.The intestinal propulsion rate of the model group was higher than the EA sensitized acupoint group,EA non-sensitized acupoint group and the blank group after electroacupuncture for 7 days.The difference was statistically significant(P < 0.05).There was significant difference in EA non-sensitized acupoint group compared with normal group and EA sensitized acupoint group(P < 0.05).And no significant difference between EA sensitized acupoint group and normal group.4.The colon length of the model group was lower than EA sensitized acupoint group point group,EA non-sensitized acupoint group and normal group after 7 days of treatment,and the difference was statistically significant(P < 0.05).The EA non-sensitized acupoint group was different from the normal group and the EA sensitized acupoint group(P < 0.05).There was no significant difference between the EA sensitized acupoint group and the normal group in colon length.5.The A1 R and A3 R in the colon of the model group were significantly lower than normal group after the treatment,(P < 0.05).The A1 R and A3 R in the colon of the EA sensitized point group and the EA non-sensitized acupoint group were higher than model group,the difference was statistically significant(P < 0.05).Compared with EA non-sensitized acupoint group,A1 R and A3 R in the colon of EA sensitized acupoint group was statistically significant(P < 0.05).Conclusion:1.TNBS(2,4,6 three nitrobenzene sulfonic acid)-ethanol method can copy the intestinal motility in the model of inflammatory bowel disease effectively.2.EA can be used to treat the symptoms of hyperactivity of stomach and intestines by improving intestinal motility in model mice with IBD,and the curative effect of EA sensitized point acupoints is more significant compared with non-sensitized acupoints by EA.3.EA sensitized acupoint may inhibit the intestinal motility of IBD model by increasing the content of A1 R and A3 R receptors in the colon,thereby improving the symptoms of intestinal hyperactivity in IBD mice.4.The effect of EA sensitized acupoint on disordered intestinal motility in IBD model mice is better than EA non-sensitized acupoint.EA is safe and effective for the treatment of IBD,It has a good effect on the intestinal motility in IBD model mice,which is worthy of clinical reference.
