Objective:To observe the effect of Dendrobium Compound(DC)on the inflammatory factors in the kidney of diabetic nephropathy mice,and to investigate the possible mechanism of DC in the treatment of DN,providing new ideas and new targets for the prevention and treatment of DN by traditional Chinese Medicine.Methods:After a one-week adaptive feeding,32 male db/db mice(body weight about 45g,SPF level,11?12 weeks of age),were randomly divided into 4 groups:model group,positive control group,low and high dose of DC groups,and each group has 8 mice.In addition,8 normal db/m mice of the same age were selected as normal control group.The equivalent dose was calculated as the ratio of human body surface area to the body surface area of the mouse,and the animals in each group were treated with intragastric administration respectively,two DC treatment groups of db/db mice were administered with DC at doses of 6g/(kg·d)and 12g/(kg·d)respectively,the positive control group was given Glipizide 2mg/(kg·d),and the normal control group and the model group were administered with normal saline of equal capacity.The administration of each group was performed by gavage daily for a period of 8 weeks.The body weight of mice was measured every 2 weeks during treatment.At the beginning of the experiment and the end of the fourth and eighth weeks,the blood was collected by cutting the tail of the mouse,and fasting blood glucose was measured rapidly in each group by blood glucose meter.Oral glucose tolerance test(OGTT)was performed one day before the end of the experiment.At the end of the experiment,all the mice were killed and serum related biochemical indexes were detected:glycosylated serum protein(GSP),insulin(Ins),total cholesterol(TC),triglyceride(TG),serum creatinine(Scr),blood urea nitrogen(BUN);HE and PAS staining were used to observe the pathological changes of renal tissue;ELISA was used to detect the protein expression of tumor necrosis factor alpha(TNF-a),monocyte chemoattractant protein-1(MCP-1),intercellular adhesion molecule-1(ICAM-1)and interleukin-6(IL-6)in serum and kidney tissues.Western blot was used to detect the protein expression of TNF-?,MCP-1,ICAM-1 and IL-6 in renal tissue.Results:1 The condition of mice in each group:At the beginning of the experiment,compared with the normal group,the mice in the model group are obviously polydipsia,polyphagia,polyuria and obese.Their fasting blood glucose measured significantly high(P<0.01).After the drug treatment,the above situation better than before.2 FBG?GSP?Ins:Compared with the normal group,FBG?GSP and Ins in the model group increased significantly(P<0.01);compared with the model group,FBG?GSP and Ins in DC treatment groups and the positive control group decreased significantly(P<0.05,P<0.01);DC treatment groups compared with the positive control group,the difference was not statistically significant(P>0.05).3 OGTT:FBG in the model group was significantly higher than that of the normal group(P<0.01),Blood glucose increased significantly after oral administration of glucose for half an hour later(P<0.01),blood glucose decreased slowly after 60 and 120 minutes(P<0.01).FBG in DC treatment groups and the positive control group was significantly lower than that of the model group(P<0.01),and the level of blood glucose was lower than that of the model group after oral administration of glucose for half an hour later(P<0.05),then it decreased continuously after 60 and 120 minutes(P<0.01).DC treatment groups compared with the positive control group,the difference was not statistically significant(P>0.05).4 TG?TC:Compared with the normal group,TG and TC in the model group increased significantly(P<0.01).Compared with the model group,TG and TC in the positive control group decreased significantly(P<0.05,P<0.01);TG and TC in the low dose of DC group reduced,but the difference was not statistically significant(P>0.05);while TG and TC in the high dose of DC group reduced significantly(P<0.01).5 Scr?BUN:Scr and BUN in the model group were significantly higher than that of the normal group(P<0.01);Scr and BUN in DC treatment groups and the positive control group were significantly lower than that of the model group(P<0.05,P<0.01);there were no significant difference between DC treatment groups and the positive control group(P>0.05).6 Pathological changes of renal tissue:Glomerulu hypertrophy of the mice in the model group was obvious,and their mesangial region widen,their capillary basementmembrane thickened;both DC treatment groups and the positive control group were better than the model group;the degree of renal disease gradually improved with the increase of dosage of DC.7 Protein expression of TNF-a,MCP-1,ICAM-1 and IL-6 in serum and kidney tissue of mice:Compared with the normal group,TNF-a,MCP-1,ICAM-1 and IL-6 in the model group increased significantly(P<0.01);compared with the model group,TNF-?,MCP-1,ICAM-1 and IL-6 in DC treatment groups and the positive control group decreased significantly(P<0.05,P<0.01).Conclusion:1 DC can reduce FBQGSP and hyperinsulinemia;significantly improve blood lipid levels,the renal function and pathological damage.2 DC has protective effect on the kidney of diabetic nephropathy db/db mice.It may delay the progress of diabetic nephropathy by reducing the expression level of TNF-?,MCP-1,ICAM-1 and IL-6,and inhibiting the inflammatory reaction.
