Objective: By observing the effect of water extract of Condonopsis lanceolata on the signal pathway of AMPK / PGC-1? in skeletal muscles of exercise-induced fatigue rats,the possible mechanism of anti-exercise fatigue of Condonopsis lanceolata's water extract was discussed,which provided experimental basis for the clinical treatment of exercise-induced fatigue.Method: 64 male SD rats were randomly divided into 4 groups: normal group,model group,Condonopsis lanceolata group and positive control group,with 16 rats in each group.The normal group and the model group were given the same dose of distilled water daily,and the positive control group was treated with Rhodiola capsules as a comparison of efficacy.In addition to the normal group,rats were subjected to weight-bearing swimming training 1 hour after the daily gavage to establish an exercise-induced fatigue model.After 4 weeks of weight-bearing swimming training,one-time exhaustion exercise was performed and the exhaustion time of each group of rats was recorded.Immediately after exhaustion,8 rats in each group were selected and serum,liver,and gastrocnemius muscle samples were prepared after anesthesia;12 hours after exhaustion,the other half of the rats in each group were anesthetized and a gastrocnemius muscle tissue sample was prepared.HE staining was used to observe the pathological changes of rat gastrocnemius muscle.Serum CK,BLA,BUN and liver LG,SOD and MDA levels were measured by kits to explore the mechanism of energy metabolism and oxidation.Finally,the expression of AMPK and PGC-1? protein in gastrocnemius muscle was measured by Western blot immediately after exhaustion and after 12 hours of exhaustion.From shallow to deep,the biological mechanism of exercise-induced fatigue and the molecular targets of the water extract of Condonopsis lanceolata were analyzed layer by layer.Result:(1)Compared with the model group,the rats in the Condonopsis lanceolata group and the positive control group both significantly prolonged exhaustion swimming time(P<0.01);(2)the liver LG and SOD contents in the Condonopsis lanceolata group and the positive control group were higher than those of the model.The serum levels of CK,BLA and BUN were significantly lower than those in the model group(P<0.05);(3)Immediately after exhaustion: AMPK and PGC-l? protein in the model group were significantly higher than those in the normal group(P<0.01).Compared with the model group,the expression of AMPK and PGC-l? protein in the Condonopsis lanceolata group and the positive control group were significant difference(P<0.05);(4)After 12 hours of exhaustion: Expression of AMPK and PGC-l? protein in the skeletal muscles of the model group was significantly reduced immediately after exhaustive(P<0.01).After treatment with the drug,the Condonopsis lanceolata group and the positive control group AMPK,PGC-l? protein was still significantly higher than the model group(P <0.01).Conclusion: The water extract of Condonopsis lanceolata can effectively treat exercise-induced fatigue,and its mechanism may be to promote the high expression of AMPK and PGC-l? protein in skeletal muscle,thereby increasing the hepatic glycogen reserve and SOD activity,thereby alleviating skeletal muscle exercise fatigue.
