ObjectiveTo observe the therapeutic effect of Bushen Jiedu Decoction?BJD?on reproductive organ and reproductive related cell injury in mice induced by ionizing radiation,and to explore the protective effect of this decoction on the reproductive system injury caused by ionizing radiation,combined with the treatment theory of nourishing Yin and replenishing essence,clearing away heat and toxic material in traditional Chinese medicine.MethodsThe mice of genetic background C57BL/6J,Female rats were respectively and randomly divided into 3 groups:blank control group of female?A1?,radiation model group of female?A2?,the BJD group of female?A3?.And male rats were respectively and randomly divided into 3 groups:blank control group of male?B1?,radiation model group of male?B2?,the BJD group of male?B3?.It was divided into 6 groups.Each group has 20 mice.The A3 group and B3group was given BJD?13.5/kg·d?by gavage.The other groups were given were 0.2ml of physiological saline by gavage.Each group was treated for 24days.Except for the A1 group and B1 group,the other four groups received the whole body 5 Gy 60Co?ray radiation for one time on the eleventh day.The general physical signs of mice in each group were observed.On the first day and the fourteenth day after radiation,the weight of mice was measured and the testis index,ovary index were calculated.The pathomorphological changes of testis,epididymis,ovary and uterus were observed on the fourteenth day with light microscope.The number of sperm in epididymis was counted,the rate of sperm deformity and survival rate were measured,and the number of follicles at different levels in the ovary was counted under optical microscope on the fourteenth day.Results?1?The organ index of reproductive organs:?1?Testis index:First day after radiation,compared with group A1,the testis index in group A2 and A3 was significantly decreased?P<0.01?,but in group A3 was higher than that in group A2?P<0.05?;At fourteenth day after radiation,the testis index in group A2 was significantly lower than that in group A1?P<0.05?,and the testis index in group A3 was significantly higher than that in group A2?P<0.05?,and there was no significant difference from that in A1 group?P>0.05?.Compared with the first day after radiation in the same group,the fourteenth day testis index of A3 group increased significantly?<0.05?.?2?Ovarian index:first days after radiation,compared with group B1,the ovarian index of group B2 and B3 group decreased significantly?P<0.01?,and there was no statistical difference between B2 group and B3 group?P>0.05?.At fourteenth day after radiation,the ovarian index of group B2 and group B3 was lower than that in group B1?P<0.05 or P<0.01?,and the ovarian index in group B3 was significantly higher than that in group B2?P<0.05?.The ovarian index of B3 group was significantly higher than that in the same group after first days after radiation?P<0.01?,and there was no significant difference between the other groups?P>0.05?.?2?Pathomorphology:In the A2 group and B2 group of mice,the pathomorphology of testes,epididymides,ovary,uterus showed injuries in different degrees.The tissue structure in the A3 group and B3 group of mice were basically recovery after Intervention of Chinese medicine compound.?3?Sperm count:Fourteenth day after radiation,the number of sperm in the A2group and A3 group was significantly lower than that in the group A1?P<0.05or P<0.01?,but the number of sperm in the A3 group was significantly higher than that in the A2 group?P<0.05?.?4?The rate of sperm abnormality:Fourteenth day after radiation,the rate of sperm abnormality in the A2 group was significantly higher than that in the A1 group and A3 group?P<0.05?,and there was no significant difference between the A1group and A3 group?P>0.05?.?5?Sperm survival rate:Fourteenth day after radiation,the survival rate of sperm in the A2 group was significantly lower than that in the A1 group and A3 group?P<0.05?.There was no significant difference in the survival rate of sperm in the A3 group compared with that of the A1 group?P>0.05?.?6?The follicle counts at all levels:Fourteenth day after radiation,Compared with group B1,the number of follicular follicles at different levels in group B2decreased to varying degrees?P<0.05 or P<0.01?,the number of initial follicles,secondary follicles and antral follicular follicles in the B3 group decreased significantly?P<0.05?.The number of primary follicles in group B3was not statistically significant?P>0.05?compared with that in group B1.Compared with the number of primary follicles,secondary follicles and antral follicles in group B2,the number of follicular follicles in each group of B3group increased to varying degrees?P<0.05 or P<0.01?.ConclusionsBJD can reduce the damage of reproductive system caused by exposure to ionizing radiation:?1?to reduce the pathomorphological damage of the epididymis,testis,ovary and uterus of the reproductive organs.?2?increase the testicle index of male rats,improve the sperm quality of life in epididymis,increase the number and survival rate of sperm,and reduce the incidence of abnormal sperm.?3?increase the ovaries index of female rats,and restore the number of primary follicles,secondary follicles and sinus follicles in the ovary.
