Abstract/Summary:
ObjectiveThe purpose of this study is to explore ERMP1 protein expression in pancreatic cancer and its correlation with clinical pathology and prognosis and the effect of ERMP1 on pancreatic cancer cell invasion and metastasis.Our study has important theoretical guiding significance and clinical application value for improving the curative effect of pancreatic cancer.Methods1.The expression of ERMP1 protein in normal pancreatic tissue and pancreatic ductal carcinoma was detected by immunohistochemistry,and statistical software was used to analyze the clinicopathological data correlation.2.Different metastatic potential pancreatic cancer cell lines Aspc-1,Panc-1 and Capan-2(high/medium/low)were cultured in normoxia and hypoxia(oxygen partial pressure <1%,48h).The transcription and protein expression levels of ERMP1 in three pancreatic cancer cells under different culture conditions were then examined by real-time quantitative PCR and Western blot.3.The ERMP1 knockout stable cell line was constructed in Panc-1 cells using the CRISPR/Cas9 gene editing system to explore the effects of ERMP1 down-regulation on the invasion and metastasis of pancreatic cancer cells under hypoxia.Results1.According to the results of immunohistochemistry experiments,we found that ERMP1 protein was highly expressed in PDAC tissues,and was present in normal pancreatic tissues with low or no expression.and ERMP1 expression was significantly associated with TNM stage and lymph node involvement,the clinical prognosis was also statistically significant.2.Under the condition of hypoxia and normoxic culture,we found that the invasion and metastasis capacity of pancreatic cancer Panc-1 cells cultured under hypoxia was stronger than that of cultured under normoxia;Besides,the invasion and metastasis capacity of pancreatic cancer Panc-1 cells decreased significantly after downregulation of ERMP1 under different culture conditions.Conclusions1.The unusual expression of ERMP1 in PDAC tissues is related to the biological behavior of PDAC,especially lymph node metastasis and TNM staging.2.The capability of invasion and metastasis of pancreatic cancer cells were enhanced under hypoxic conditions:The invasion and metastasis capacity of pancreatic cancer Panc-1 cells was down-regulated after down-regulation of ERMP1,suggesting that ERMP1 may have some influence on the invasion and metastasis of pancreatic cancer cells.
