ObjectiveTo investigate the main role of LAMP2 a in colorectal cancer,its expression and clinicopathological significance.Methods1.Checking the LAMP2 a m RNA of colorectal cancer and normal mucosa by RT-PCR.2.Using Western blot to test the expression level of LAMP2 a protein in colorectal cancer and normal mucosa.3.Using laser capture microdissection(LCM)to identify the expression of LAMP2 a m RNA in colorectal adenocarcinoma lesions and normal glandular epithelium.4.Construct tissue chip and using immunohistochemistry to test LAMP2 a of colorectal cancer(n=374),normal mucosa(n=367)and adenoma tissues(n=18),and analysis clinicopathological features of LAMP2 a expression in colorectal cancer.5.The bioinformatics analysis of LAMP2 mRNA expression in colorectal cancer.6.We use SPSS 17.0 software to test all data.statistical software to analyze data.Data were expressed as meanąstandard deviation,differences using independent sample T-test to analyze the differences,Spearman rank analysis was used to analyze the relationship between different pathological features and LAMP2 a in colorectal cancer.Kaplan-Meier survival plots were generated.The Cox proportional hazards model was employed for multivariate analysis.Results1.The expression of LAMP2 a m RNA and protein in colorectal cancer tissue.RT-PCR results showed that LAMP2 a mRNA was lower expressed in colorectal cancer than normal mucosa(P<0.05).The expression of LAMP2 a m RNA was analyzed by laser capture microcutting technology(LCM).The results showed that the expression of LAMP2 a was lower in colorectal cancer than normal mucosa.The expression of LAMP2 a protein was higher in colorectal cancer than normal mucosa(P<0.05).2.The LAMP2 a protein expression in colorectal cancer tissue and its relationship with clicopathological features.LAMP2a expression was higher in colorectal cancer and adenoma than normal mucosa(P<0.05),LAMP2 a expression was negatively correlated with TNM stages and lymph node metastasis(P<0.05),but was no relationship with lymphatic invasion,liver metastasis,depth of invasion and differentiation(P>0.05).Kaplan-Meier analysis showed that LAMP2 a expression was not linked to favorable prognosis of colorectal cancers(P>0.05).Cox proportional hazard model showed that depth of invosion and distant metastasis were independent prognostic factors of colorectal cancer(P<0.05).3.According to the data of Skrzypczak2 and TCGA,the expression of LAMP2 mRNA was higher in colon tissue than adenomas tissue,and the same result was found in the rectum tissue.TCGA datas also showed that LAMP2 m RNA expression was higher in the colon tissue than rectal tissue.The patients of MSI and K-ras LAMP2 expression were higher in patients of negative MSI and K-ras than positive patients.The expression of LAMP2 was higher in patients with normal BRAF gene expression than the patients with abnormal BRAF gene.Conclusions1.The expression of LAMP2 a mRNA was lower in colorectal cancer tissues than normal mucosa tissues,while the protein expression was higher than normal mucosa tissues.2.Clinicopathological analysis showed that LAMP2 a expression was negatively correlated with TNM stage and lymph node metastasis in colorectal cancer.Kaplan-Meier analysis showed that LAMP2 a expression was not linked to favorable prognosis of colorectal cancers.3.Cox proportional hazard model showed that depth of invosion and distant metastasis were independent prognostic factors of colorectal cancer...
