ObjectiveTo study the inhibitory effect and the action mechanism of miR-101 on the proliferation and invasion of retinoblastoma.MethodsThe cells of HXO-Rb44,miR-101 mimics and miR-101 negative control were cultured,and the experiment included three groups such as miR-101expression group,MiR-101 negative control group and normal HXO-Rb44cells as blank group.The miR-101 mimics were detected by reverse transcription-polymerase chain reaction?RT-PCR?and western blot MiR-101.Histone-lysine N-methyltransferase?Histone-lysine N-methyltransferase?were detected in miR-101 negative control group and retinoblastoma cell HXO-Rb44blank control group EZH2)expression.The proliferation and invasion ability of cells in each group were determined by MTT assay and Transwell assay.Finally,luciferase reporter assay was used to determine the target relationship between miR-101 and EZH2.Resultshe relative expression level of miR-101 in miR-101 group was significantly higher than that in blank group and negative control group?P<0.05?.The relative expression levels of EZH2 in blank group and negative control group were significantly higher than that in miR-101 group?P<0.05?.The OD49090 value of EZH2 in miR-101 group was significantly lower than that in blank group and negative control group?P<0.05?The number of EZH2-penetrating membrane in the group and the negative control group were significantly higher than that in the miR-101 group?P<0.05?.EZH2 is the target of miR-101.ConclusionsUp-regulation of miR-101 inhibited the proliferation and invasion of HXO-Rb44 cells,and its mechanism of action was related to EZH2.
