ObjectiveInvestigate the difference of Septin2 gene expression in gastric normal tissue and gastric carcinoma and its effect on proliferation and apoptosis of gastric cancer cells.MethodsThe m RNA expression of Septin2 in gastric cancer tissues and normal gastric tissues from GSE54129 data set of GEO database was compared.The expression of Septin2 at protein level was analyzed by immunohistochemical staining of gastric normal tissues and gastric cancer tissues.Survival analysis was performed using the Kaplan-Meier Plotter database.The SCG-7901 and MNK-45 cell lines were transfected with the plasmid containing Septin2 gene to upregulate Septin2 expression and then cell proliferation and apoptosis were analyzed.ResultsThe expression of Septin2 gene in gastric cancer tissue at m RNA level(P <0.001)and protein level(P <0.001)was significantly higher than that of normal tissue.Septin2 expression was associated with prognosis and survival time of patients with Septin2 high expression was shorter than that of low expression(Logrank P < 0.001).Overexpression of Septin2 promotes proliferation(P<0.001)and inhibits apoptosis(P<0.001)in gastric cancer cells.ConclusionsThe expression of Septin2 in gastric cancer tissues is higher than that in normal tissues,which is closely related to the occurrence and prognosis of gastric cancer.The overexpression of Septin2 gene can promote the proliferation and inhibit its apoptosis.
