Objective:To study the impact of eclipta on the characeristics of melanocytes,including the phosphorylation levels of ERK1/2 and p38,and MITF expressions,which can understand the mechanism of its role.Methods:Use rat immortalized melanocytes(B10BR);Set different concentrations of eclipta;Established the blank control group and using antibody inhibitor as positive control group.The expressing of MITF and the phosphorylation levels of ERK1/2 and p38 was measured by Western Blot.Results:According to the one-way analysis of variance,the dosage of 50?100?200?g/mL eclipta significantly increased the phosphorylation levels of ERK1/2.The phosphorylation levels of ERK1/2 in positive control group with ERK1/2 inhibitor was significantly inhibited;The dosage of 50?100?200?g/mL eclipta significantly increased the phosphorylation levels of P38.The phosphorylation levels of p3 8 in positive control group with p3 8 inhibitor was significantly inhibited;the dosage of 50?100?200?g/mL eclipta significantly increased the expressing of MITF.The expressing of MITF in both positive control groups was significantly inhibited.Conclusions:(1)Eclipta can increase the phosphorylation levels of p38 and ERK1/2.(2)Eclipta can increase the expressing of MITF.(3)Eclipta may increase the expressing of MITF by the way of increase the phosphorylation levels of p38 and ERK1/2,which ultimately affect the TYR and melanocytes.
