Objective:Observing the effect of using external application with Xugudan on expression of vascular endothelial growth factor(VEGF)and transforming growth factor-?(TGF-?)in rat model of tibial nonunion.Methods:A total of 10 SD rats as blank group,model 50 SD rats were randomly divided into model group,high and low dose Xugudan(150,75,25mg,kg-1)groups,transcutaneous electrical nerve stimulation(TENS)group.In 4 weeks,8 weeks after the intervention of rats for X-ray detection and measurement of bone spacing;hematoxylin eosin(HE)staining was used to observe the morphological changes;Real-time PCR and enzyme linked immunosorbent assay(ELISA)were detected in bone injury the serum VEGF,TGF-? changes.Results:X-ray examination:4 weeks after intervention,the bone scab began to grow in the nonunion area except the model group.After 8 weeks of intervention,the fracture line disappeared and the bone cortex was continuous,and the bone space was continuously measured.Compared with the model group,the interval of bone in each group of rats decreased significantly(P<0.01),and the phase of group TENS was compared with that of the group.In the 4 and 8 weeks after intervention,the healing degree of bone space in the high dose group was faster(P<0.01).There was no significant difference between the bone distance in the dose group of Xugudan,and the degree of bone distance healing was slow in the low dose group of Xugudan(P<0.01).Histomorphological detection:4 weeks after intervention,the bone trabecula appeared in each group,but the bone cortex was not continuous.8 weeks after intervention,the bone trabecular bridge was connected in each group and the bone cortex of the continuous bone high dose group was continuous.The content of VEGF and TGF-? in the experimental group was higher than that of the model group(P<0.01)except for the blank group.After the treatment of Chinese medicine,the content of VEGF and TGF-? in the Xugudan group increased with the increase of dose(P<0.01).Compared with the TENS group,there was no significant difference in the dose group of the Xugudan,and there was a significant difference between the continued bone and low dose groups(P<0.05,P<0.01).Conclusion:Xugudan can improve the local blood supply by regulating the expression of VEGF and TGF-?,induce vascular remodeling and bone remodeling,and promote the nonunion healing.
