Objective:To investigate the expression of key molecules PTEN,PI3K,AKT and mTOR in mTOR signaling pathway of keloids and normal skin,and whether there is an effect of Gallnut scar ointment on mTOR signaling pathway and to clarify the target of Galla chinensis cream.Research content and methodsIn this experiment,keloid fibroblasts as the research object,the human keloid tissue transplanted on the back of the nude mice to mold,to smear Galla chinensis cream as experimental group,smear matrix as a control group,and then normal skin tissue as a blank control group The expression of PTEN,P13K,Akt,and mTOR,the key molecules of the mTOR signaling pathway,was studied by immunohistochemistry to discriminate the differences among the three groups,a.nd the mechanism and target of the detoxification of phlegm,soft and firm were defined.Six-week-old nude mice(BALB/C-nunu strain),females,weighing 18-22 g,were purchased from the Experimental Animal Center of Xuzhou Medical University and 37 batches of four batches were adapted for 1 week under sterile conditions.Under strict aseptic conditions,the surgical incised human pre-thoracic scar was cut into 5mm x.7mm x 3mm tissue pieces,and the keloid tissue pieces were implanted subcutaneously in the back of nude mice.After about 16 days,a living keloid was made.Animal model.Eighteen female BALB/C strain nude mice were selected in good condition.Nude mice were randomly divided into 2 groups,control group and experimental group,with 9 mice in each group.The experimental group and the control group started to use drugs when they were wounded.The experimental group was smeared with gall nut cream,and the control group was only coated with the matrix(matrix made of scar cream).Observe the condi tion of the keloid implanted every day.Apply Gallnut Cream and Substrates to the Scar3 times/day,the nude mice wore executed after 28 days,and the fresh prepuce tissue was taken as blank control in the skin surgery of the central hospital of xuzhou city.The three groups of tissues were stained with HE,and the immunohistochemical.SP method was used to detect the changes of PTEN,P13K,Akt and mTOR,and the photo record was saved,and the data statistical analysis was performed using the IBM SPSS20.0 software package.The results of the study1 PI3K mainly in keloid fibroblasts cytoplasm and small nucleus expression,tan,gallnut scar cream treatment group(33.33%)and lower the expression(40.00%),normal skin scar tissue matrix control group(77.78%)increased,the difference was statistically significant(P<0.05).2.The main expression of PTEN in keloid in fibroblast nuclei with a small amount of cytoplasm,tan,keloids in the matrix control group(22.22%)of the middle and lower expression,gallnut scar cream treatment group(88.89%)and has a high expression in normal skin(100.00%),the difference was statistical ly significant(P<0.05),3.MTOR in keloid main expression in f ibroblasts cytoplasm and smnall nucleus,tan,gallnut scar cream treatment group(11.11%)and lower the expression(00.00%),normal skin scar tissue matrix control group(77.78%)increased,the difference was statistically significant(P<0.05).3.Akt in keloid main expression in fibroblast nuclei with a small amount of cytoplasm,tan,keloids in the matrix control group(100.00%)increased,gallnut scar cream treatment group(33.33%)and with lower expression in normal skin(60.00%),the difference was statistically significant(P<0.05),4.Correlation analysis of Person showed that PTEN in keloid was negatively correlated with PI3K,mTOR and Akt(P<0.05),and PI3K was positively correlated with mTOR and Akt(P<0.05),and Akt was positively correlated with mTOR(P<0.05).Conclusion1.Keloid fibroblasts mTOR signaling pathways in the abnormal expression,PTEN and PI3K,and AKT,mTOR is one of the key molecules,by gallnut scar cream after the intervention,PI3K,and AKT in keloid fibroblasts,mTOR expression is restrained,PTEN expression,expression levels close to normal skin tissue,suggesting that the curative effect of Chinese gall scar cream in the treatment of keloid,exactly through the mTOR signaling effect of intervention of scar fibroblasts proliferation can confirm that the signal of drug targets for PTEN in signaling pathways,P13K,and AKT,mTOR inhibitors.To provide a theoretical basis for the follow-up study on the treatment of keloid in keloid.
