Objective: To observe the effects of Dian sanguis draconis on experimental pulmonary fibrosis in rats.To observe the effects of Dian sanguis draconis on the dynamic expression of IL-4/IFN-? in serum and the level of FIZZ1 in lung tissue of rats.To investigate the mechanism of action of Dian sanguis draconis on pulmonary fibrosis.Methods: Rats pulmonary fibrosis model was prepared by intratracheal bleomycin infusion(5mg / kg).Ninety-six male Sprague-Dawley(SD)rats were randomly divided into a total of 6groups--control group(group A),model group(group B),Dian sanguis draconis whole intervention group(group C),Dian sanguis draconis early intervention group(group D),Dian sanguis draconis lately intervention group(group E)and pirfenidone group(group F).The rats were orally administered with the corresponding drugs 1 day after the model was established.The levels of IL-4 / IFN-? in serum were detected by ELISA.The protein and m RNA expression of FIZZ1 in lung tissue were detected by immunohistochemistry(IHC)and Real Time PCR.Colorimetry was used to determine the content of hydroxyproline(Hyp)in lung tissue.HE staining and Masson triple staining were used to analyze the pathological morphology.Result:The early and whole intervention of Dian sanguis draconis could inhibit the expression of FIZZ1 m RNA and protein in the lungs of experimental PF rats(P <0.01),decrease serum IL-4 level(P <0.01 or P <0.05)and increase serum IFN-?(P <0.01 or P<0.05).The level of Hyp in the lungs of PF rats was decreased(P <0.01 or P <0.05).Histopathology confirmed Dian sanguis draconis can reduce the degree of pulmonary alveolitis and pulmonary fibrosis.The results showed that the overall effect of the full intervention of Dian sanguis draconis treatment group was the best,with no significant difference with pirfenidone group(P> 0.05).Conclusion:The early and whole intervention Dian sanguis draconision of can effectivelyreduce bleomycin-induced alveolitis and fibrosis.The mechanism may be relate to the regulation the imbalance of IL-4 / IFN-?,and to the inhibit the expression of FIZZ1,a core promoter of "inflammation cascade reaction" in the lung.Thereby preventing follow-up of collagen deposition,angiogenesis and other series changes.Dian sanguis draconis has a multi-target anti-inflammatory effect to the core,with in-depth research and development value in the field of early prevention and treatment of PF.
