Prostate cancer(PC)is one of the most common malignant tumors in elderly men.It can be divided into two kinds:familial and sporadic,both of which are the result of interaction between genetic factors and environmental factors.Worldwide,its incidence is the second place and the mortality rate is sixth in all malignant tumors of men.Especially in the US,the incidence of PC has exceeded lung cancer,and it has become the first cancer that endangers men's health.In Asia,the incidence of PC is much lower than that in European and American countries,but it has been increasing rapidly in recent years.Studies have shown that in 1998,the incidence of PC in China was 3.52/10 million,and the incidence increased to 212.5%in 2008,reaching 11.00/10 million.The average annual growth rate was 12.07%in 10 years.Therefore,it is of great significance to prophylaxis and treatment of PC,which we should study occurrence,development and regularity of PC deeply.According to recent literatures and data analysis,there is evidence that the occurrence and development of PC may be closely related to the OCT2(organic cation transporter member 2,SLC22A2),but there are relatively few literatures about it.Objective:In the present research,the author tries to explore the expression of OCT2 in PC tissues and opposite PC paracancerous tissues and then analyse the differences in the expression.The author would to explore the relationship between the expression of OCT2 and the clinicopathological parameters,and then analyze the role of OCT2 in the occurrence,development,invasion and metastasis of prostate cancer.The author also would to study the correlation between OCT2 and methylation.Method:118 cases of prostate cancer patients were collected from 2016 to 2017,and prostate cancer tissues and opposite paracancerous tissues were used as control.After all pathological tissues were stained with HE confirming pathological diagnosis,immunohistochemical method(IHC)expression was detected in 118 cases of prostate cancer tissues and corresponding adjacent tissues.We also performed RCR(polymerase chain reaction)and RT-qPCR(real time quantitative polyerase chain reaction)to detect the DNA levels and mRNA levels of OCT2 in another 63 cases of prostate cancer tissues and corresponding adjacent tissues.The genetic test found that OCT2 gene promoter region has CpG Island(CPI).The immunofluorescence assay was used to detect reporter gene carrier pCpGL having CpG island of the active site of the OCT2 promoter,which was added to the methylation enzyme in vitro.Statistics software SPSS 19.0 was adopt for data analysis.The P values was less than 0.05 were considered to be statistically significant.Results:1.The expression of OCT2 in prostate cancer tissues and opposite paracancerous tissues:Specifically speaking,OCT2 is mainly located in the cytoplasm,less in the cell membrane.The positive expression rates of OCT2 in the prostate tissue and the corresponding para cancer tissues are 16.9%(10/58)and 82.8%(48/58)(P<0.005).The difference in the expression rate of OCT2 in the prostate cancer tissue and the para cancer tissue is statistically significant.2.The difference in the expression rate of OCT2 in the prostate cancer tissue and the para cancer tissue is statistically significant.3.The low level expression of OCT2 in PC is associated with DNA methylation.
